目的:探讨荧光原位杂交技术(FISH)检测乳腺癌Her2/neu的扩增情况,比较FISH法与免疫组织化学(IHC)法检测结果的一致性。方法:收集乳腺癌患者手术切除的肿瘤标本50例。经石蜡包埋,组织切片后,应用FISH法检测Her2/neu扩增情况,IHC法检测Her2/neu蛋白表达情况,并对两种检测方法的结果进行统计学分析。结果:50例标本中FISH阳性17例,FISH阴性33例,FISH阳性率34%(17/50)。 50例标本中IHC(~)14例,IHC(0~+)36例,IHC蛋白表达率28%(14/50)。FISH法与IHC法检测结果的总符合率为82%(41/50)(K=0.581)。结论:FISH法具有较高的稳定性与可靠性,临床开展FISH法检测Her2/neu基因扩增情况作为IHC法的补充, 可以更加准确和客观地对乳腺癌患者的治疗和预后进行评价。
Objective To identify the expression of nerve growth factor (NGF) and its high affinity receptor (tyrosine kinase receptor A, TrkA) during bone induction by recombined human bone morphogenetic protein 2 (rhBMP-2) by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR) and to discuss the role of NGF on the bone induction of the BMP. Methods Thirty-six ICR mice were divided into the experimental groupand the control group at random. rhBMP-2 /collagen sponge and collagen sponge were implanted into the right thigh muscle pouches of the mice in the experimental group and the control group, respectively. The tissues in the implanted site of the two groups were removed on the 7th, 14th and 21st day after the implantation. Histological, immunohistochemical and RT-PCR analyses were performed to detect osteoinductive effects of rhBMP-2 and the expression of NGF and TrkA. Results Gross observation showed that a solid lump was found in theright thigh in the experimental group on the 7th day and became harder on the 14th and 21st day, which was not found in the control group. rhBMP-2/collagen sponge displayed a potent ability to induce bone formation, while immunostaining for NGF and TrkA was observed in the course of osteoinduction by rhBMP-2. On the 7th day in the experimental group, NGF positive immunostaining reached the peak in thestage of chondrogenesis and there were a large number of cells expressing NGF, including fibroblasts, chondroblasts, chondrocytes, and osteoblasts; then, therewas a decrease in the number of the positivecells and in the intensity of immunostaining on the 14th and 21st day. Staining of TrkA wassimilar to that of NGF. The expression level of the mRNA of NGF during the course of bone induction peaked 7 days after the implantation and then decreased. Conclusion rhBMP-2/collagen is a kind of satisfactory osteoinductive material, and many different kinds of cells induced by rhBMP-2 can express NGF and TrkA, which suggests that NGF may play an important role in the osteogenesis initiated by exogenous BMP through direct and indirect pathways.
Objective To observe the difference between blood brain barrier and blood optic nerve barrier. Methods Twenty normal male SD rat sprime; optic nerve including prelaminar region, lamina cribrosa, retro-laminar region, intraorbital portion, intracanalicular portion, and intracranial portion respectively,and cerebral cortex were removed separately. Ultrastructure of endothelial cells was observed by electron microscopy. Immunohistochemical staining was used to detect the expression of transferrin receptor (OX-26) and metalloproteinase inducer (OX-47) and extravasation of fibrinogen around microvessels. Results The results of electron microscopy showed that endothelial cells of microvessels in each portion of optic nerves and cerebral cortex did share the same tight junctions. However, the number of plasmalemmal vesicles in prelaminar region was significantly more than that in cerebral cortex(P<0.05);there was no significant difference between other parts of optic nerves (lamina cribrosa, retro- laminar region, intraorbital portion, intracanalicular portion, and intracranial portion)and cerebral cortex in the number of the plasmalemmal vesicles(Pgt;0.05). By immunohistochemical staining,the endothelial cells of microvessels in the prelaminar region showed no expression of the OX-26 and OX-47,but extravasation of fibrinogen around microvessels was found; b positive expression of OX-26 and OX-47 was observed in the endothelial cells of the microvessels in other parts of optic nerves (lamina cribrosa, retro-laminar region, intraorbital portion, intracanalicular portion, and intracranial portion) andcerebral cortex, and no fibrinogen was seen aro und the microvessels. Conclusions There is a significant difference between the endothelial cells of the microvessels in prelaminar region and cerebral cortex in the ultrastructure, markers expression, and permeability, so the microvessel s in prelaminar region lacks the typical blood brain barrier characteristics.The microvessels in other parts of optic nerves (lamina cribrosa, retro-laminar region, intraorbital portion, intracanalicular portion, and intracranial portion) have blood brain barrier properties due to its similar specialties as which in cerebral cortex. Objective To study the expression of the fibronectin (FN) and beta;1 integrin (beta;1) in epiretinal membranes(ERM) of eyes with proliferative vitreoretinopathy(PVR). Methods wenty epiretinal membranes were obtained from eyes undergone vitrectomy for retinal detachment complicated with PVR and observed by immunohistochemical methods. Results Overexpression of FN and beta;1 were observed in 18 and 16 membranes respectively. Conclusion The synergism of FN and beta;1 in their action mignt be one of the important roles in the development of PVR. (Chin J Ocul Fundus Dis, 2001,17:119-121)
摘要:目的:探讨子宫内膜样腺癌组织中真核细胞翻译起始因子4E(eIF4E)的表达情况及意义。方法:应用免疫组织化学技术检测124例子宫内膜样腺癌及20例正常子宫内膜组织中eIF4E的表达情况,同时结合患者的临床病理资料进行分析。结果:子宫内膜样腺癌和正常子宫内膜组织中eIF4E的阳性表达率分别为726%和50%,差异有统计学意义(P=0000lt;005)。eIF4E的表达与肿瘤组织分化程度明显相关(P=0039lt;005),而与FIGO分期无明显相关(P=0296gt;005)。结论:过度表达的eIF4E可能作为判断子宫内膜恶变及恶性程度的分子标志。
目的:观察CyclinD1,P16在皮肤鳞状细胞癌组织中的表达,探讨CyclinD1,P16在皮肤鳞状细胞癌发生发展中的作用。方法:用免疫组织化学技术检测40例皮肤鳞癌石蜡包埋组织和10例正常皮肤组织的CyclinD1,P16表达。结果:CyclinD1在皮肤鳞癌组织中表达高于皮肤正常对照组,两者差异有显著统计学意义(Plt;0.01); P16在皮肤鳞癌组织中表达低于皮肤正常对照组,但两者差异无显著统计学意义(Pgt;0.05);CyclinD1,P16在皮肤鳞癌中表达呈正相关性(r=0.782,Plt;0.01)。结论:CyclinD1在皮肤鳞状细胞癌组织中阳性表达上调,P16在皮肤鳞状细胞癌组织中阳性表达下调,CyclinD1,P16在皮肤鳞癌中表达呈正相关。
摘要:目的:探讨PTTG的表达在非小细胞肺癌发生、发展中的作用及其与CMYC蛋白表达的关系。方法:应用免疫组化SP法检测PTTG、CMYC二种蛋白在44例非小细胞肺癌、20例肺良性病变组织和12例正常支气管粘膜上皮组织中的表达。结果:PTTG和CMYC蛋白在非小细胞肺癌组织中的表达明显高于肺良性病变组及癌旁组织,在TNM分期、淋巴结转移组间差别有统计学意义。非小细胞肺癌组织中PTTG与CMYC表达呈显著正相关。结论:提示PTTG和CMYC可能参与了非小细胞肺癌的发生和发展,可作为反映其生物学行为的指标。Abstract: Objective: To investigate the expression of PTTG and its relationship with expressions of CMYC protein in human nonsmall cell lung cancer (NSCLC).Methods: Immunohistochemical methods were applied to detect the expression of PTTG,CMYCproteins in 44 surgical specimens from NSCLC patients,20 pneumonic benign lesion and 19 normal bronchial epithelium. Results:There were high erexpressions of PTTG,CMYC in NSCLC tissues than inadjacent tissues and benign lesions.There were statistical relationships between their expressions and TNM stage,lymphnode metastasis.The expression of PTTG was positively correlated with CMYC. Conclusion: Overexpression of PTTG,CMYC may be related to human NSCLC,PTTG and CMYC play a cooperative role inthe process of NSCLC,all of them may be used as important indices for biologic behavior of NSCLC.
摘要:目的:研究垂体瘤转化基因在非小细胞肺癌组织、肺良性病变组织和正常支气管黏膜上皮组织中的表达, 初步探讨其与非小细胞肺癌发生发展,侵袭转移的关系。方法:(1)用SP免疫组化法检测76例临床石蜡组织标本(44例非小细胞肺癌、20例肺良性病变组织和12例正常支气管黏膜上皮组织)中的PTTG蛋白的表达。(2)用RTPCR法分析PTTG mRNA在不同性质肺组织中的表达情况。〖HTH〗结果〖HTSS〗:(1)PTTG蛋白在不同性质肺组织中的表达差别具有明显差异;在TNM分期、淋巴结转移组间差别有统计学意义。(2)PTTG mRNA在不同性质肺组织中的表达差别具有明显差异(Plt;0.001);在TNM分期、淋巴结转移组间差别有统计学意义。结论:细胞肺癌的发生发展及转移有关。Abstract: Objective: To investigate the association between PTTG expression and biological behaviors of human nonsmall cell lung cancer (NSCLC).Methods:SP immunohistochemistry was used to detect the expression of PTTG in 76 cases of paraffinembedded specimens (including 44 surgical specimens from NSCLC patients, 20 pneumonic benign lesion and 19 normal bronchial epithelium).Realtime RTPCR was performed to detect PTTGm RNA expression in 44 cases of fresh carcinoma and corresponding adjacent normal mucosa specimens.Results: The expression levels of PTTG was significantly different between normal mucosa and adenoma tissues.There were statistical relationships between their expressions and TNM stage, lymphnode metastasis. Conclusion: PTTG may play an important role in carcinogenesis、development and metastasis of human nonsmall cell lung cancer.
