Objective To investigate the value of CT grading diagnosis in clinical therapy of acute pancreatitis. Methods CT scanning was carried out in patients with acute pancreatitis between 2003 and 2009, and Enhancement CT scanning was carried out in patients without contraindications of contrast medium. Two radiologists analyzed the images and made CT grading diagnose. Contrast study was made between CT grading diagnose and clinical scale as well as prognosis. Results In 518 cases, the CT grading was as the following: grade A 9 cases, grade B 66 cases, grade C 105 cases, grade D 147 cases, and grade E 191 cases. Patients with grade A and B were fully recovered after conservation and supportive treatment. In cases of grade C, only 2 patients had recurred pancreatitis after conservation treatment, and others were fully healing. Ninety-four patients who needed operation and 23 patients who died were all occurred in grade D and E. Conclusion CT grading diagnosis of acute pancreatitis can reflect the range, extent, and course of disease, and it has good dependablity with clinical grading.
Objective Non-small cell lung cancer ( NSCLC) cells are relatively resistant to chemotherapy, and the outcomes are not always satisfactory. This study was designed to explore the relationship between the content of Ku80 protein of human lung cancer cells and their sensitivity to cisplatin.Methods The lung cancer cells isolated frommalignant pleural effusion samples frompatients with primary lung cancer were cultured in vitro. The sensitivity to cisplatin was tested with the method of CCK-8 expressed as half maximal inhibitory concentration ( IC50 ) . The relative content of Ku80 protein was determined by Western blot. The correlation between sensitivity to cisplatin of lung cancer cells and the relative content of Ku80 protein was analyzed. Results The IC50 of NSCLC group was significantly higher than that of SCLCgroup [ ( 4. 40 ±3. 39) mg/L vs. ( 1. 02 ±0. 54) mg/L, P lt; 0. 001] . The relative content of Ku80 protein of NSCLC group was statistically higher than that of SCLC group [ ( 0. 80 ±0. 45) vs. ( 0. 48 ±0. 25) , P lt;0. 05] . The correlation coefficient between content of Ku80 protein and IC50 was 0. 618 ( P lt; 0. 001) .Conclusions The content of Ku80 protein of NSCLC patients is higher than that of SCLC patients. Itmay be one of the mechanisms contributing to chemotherapeutic resistance of NSCLC. There is a negative relationship between Ku80 protein content of cancer cells and their sensitivity to cisplatin suggesting that the content of Ku80 protein may be served as a candidate index for predicting sensitivity of lung cancer cells to cisplatin.
目的:通过CT影像资料评价新生儿缺氧缺血性脑病与颅内出血量的关系及其预后。方法:收集1998~2006年临床诊断为缺氧缺血性脑病70例新生儿患者的CT资料,观察CT图像显示的脑出血量,分析不同程度的缺氧缺血性脑病和出血部位与颅内出血量和预后的关系。结果:新生儿缺氧后轻、中度出血在1~2周内均可完全吸收,发生在基底节或大脑白质区域的重度出血预后较差。结论:颅内出血量与窒息缺氧程度呈正相关;发生在不同部位的出血,其预后不同。
Objective To investigate the radiation-sensitizing effects of Ku80 silencing by siRNA interference for A549 lung cancer cells. Methods The sequences of Ku80 siRNA and negative siRNA were chemically synthesized and transfected into A549 lung cancer cells by lipofectamine. RT-PCR and Western bolt analysis were used to determine Ku80 gene expression. The transfected cells in culture dishes were irradiated with X ray at doses of 2 Gy, 4 Gy, 6 Gy, 8 Gy, 10 Gy, respectively. Once all treatments were completed, the cells were processed with the colony formation assay. Results RT-PCR detection showed that Ku80 mRNA levels in A549 lung cancer cells were reduced after transfected with Ku80 siRNA at 24 h, 48 h and 72 h time points. Western blot analysis showed that Ku80 protein content decreased at 48 h and 72 h time points compared with the control group ( P lt; 0. 05 ) . Cloning formation assay indicated that radiosensitivity of A549 lung cancer cells was enhanced after transfected with Ku80 siRNA. Conclusion Ku80 siRNA can effectively inhibit Ku80 gene expression of A549 lung cancer cells, and therefore enhance its radiosensitivity.
Objective To determine the airway wall thickness at the segmental and subsegmental levels in patients with bronchial asthma and eosinophilic bronchitis ( EB) by high resolution CT scanning,and evaluate its relationship with airway hyperresponsiveness. Methods High resolution CT scanning was performed in 14 subjects with asthma,15 subjects with EB, 15 subjects with cough variant asthma ( CVA) ,and 14 healthy volunteers. Total airway and lumen diameter, total airway cross sectional area and lumen area which corrected by body surface area ( BSA) were measured. The percentage of airway wall area to total airway cross sectional area ( WA% ) and wall thickness to airway diameter ratio ( T/D) were calculated for the right upper lobe apical segmental bronchus ( RB1) and all airways clearly visualized with a transverse diameter of 1-6 mm. Results T/D/BSA and WA% in the asthma patients were all significantly higher than those in the subjects with EB, CVA and healthy volunteers. T/D/BSA and WA% in the EB patients were significantly higher than the healthy volunteers, and similar with the CVA patients. Al /BSA in the patientswith asthma and CVA was less than the subjects with EB and the healthy volunteers. However, Al /BSA in the EB patients was similar with the healthy volunteers. Conclusions The airway wall thickness and remodeling can be measured and assessed by high resolution CT. Airway wall thickness and remodeling inEB patients are milder than asthma patients, which may be associated with airway hyperresponsiveness that presents in asthma but not in EB.
