  • Expression and Significance of Livin in Human Cholangiocarcinoma Tissues and Cell Line

    目的:探讨Livin基因在人胆管癌组织及胆管癌细胞系中的表达情况及其与胆管癌发 生发展之间的关系。方法:采用免疫组织化学技术(SP法)检测Livin基因蛋白在45例人胆管 癌标本及及40例癌旁胆管组织、20例正常胆管组织标本中的表达;同时采用RTPCR法及SP 法检测了Livin基因mRNA和蛋白在人胆管癌细胞系QBC939及非肿瘤细胞系HT1080中表达。 结果:Livin在胆管癌组织中表达阳性率为57.8%,而癌旁胆管组织、非癌胆管组织中未能检 测到Livin表达。Livin表达与性别、年龄、肿瘤大小及肿瘤分化程度无关。在有淋巴结转组 中,Livin阳性表达率(70.4%)明显高于无淋巴结转移组(38.9%)。在人胆管癌细胞QBC939 中,Livin mRNA及蛋白均特异性表达,而非肿瘤细胞系HT1080未见Livin表达。结论:Livin 基因在人类胆管癌组织和细胞系中选择性高表达,其可能与胆管癌发生、发展及预后密切相 关。

  • EXPRESSIONS OF Cbfa1 AND Osterix IN OSTEOBLASTS ON HUMAN ACELLULAR AMNIOTIC MEMBRANE

    Objective To study the differentiation of the human osteoblasts during the construction of the tissue engineered periosteum with the human acellular amniotic membrane(HAAM).Methods To construct the tissue engineered periosteum (n=60) with HAAM, the human fetal osteoblasts were used. The fetal osteoblasts were cultured for 2, 4, 6, 8, and10 days, and then their total RNA was extracted, which were reversely transcripted to cDNA. The realtime PCR analysis was used to reveal Cbfal and Osterix, and the cycle threshold (Ct) was also measured. The simplycultured osteoblasts were used as the control group (n=20).Results The expression of Cbfa1 was higher in the experimental group on the 2nd day when compared with that on the 4th, 6th, and 8th day(P<0.05). The same result existed on the 10th day when compared with that on the 4th and 8th day. The expression of Osterix increased and was highest on the 8th day when compared with the other results(P<0.05). Both of the 2 gene expressions were decreased in the control group when compared with those in the experimental group, but with no significant difference(P>0.05). Conclusion Cbfa1 and Osterix can be normally expressed by the osteoblasts after their integration with HAAM. As a scaffold, HAAM can be used to keep the osteoblast phenotype and differentiation with an osteoconductive ability. Such a cell-scaffold complex may provide a basis for the osteogenesis.

  • THE STUDY OF INTERACTION BETWEEN THE OSTEOBLAST AND BIO-DERIVED MATERIAL BY DETECTING THE GENE EXPRESSION

    Objective To study the gene expressions of human osteoblasts during the construction of tissue engineered bone with the bioderived material. Methods The fetal osteoblasts were used to construct tissue engineered bone with the bio-derived material and then were cultured 2,4,6,8 and 10 days in vitro. Real-time PCR analysis indicated that Cbfa 1, Osterix, Collagen type Ⅰ,osteocalcin(OC) and Integrin α5 and β1 were present in osteoblasts with bio-derived materials.Results The change ofCbfa1 was consistent with the change of Osterix. On 2nd day and 8th day, the expression of Osterix in experimental group was higher than that in control group, P<0.05. Collagen type Ⅰ’s change was consistent with change of OC expression, and its expression was higher in experimental group than that in control group on 2nd, 4th, 6th and 8th day. The Integrinexpression was high all along. Conclusion The important genes can be expressed normally by integrating osteoblasts with bioderived scaffolds. As skeleton tissue engineering scaffold, the bio-derived bone is conducive to keepthe osteoblast’s phenotype and differentiation with osteoconductive ability. The osteoblast can enter proliferation stage favorably and the scaffold materials exert no effects on it. Bio-derived bone can also supply more space for cellsto proliferate. The bio-derived materials promote osteoblasts adhesion.

  • Expression and significance of NgR mRNA in adult rats′ optic nerve

    Objective To observe and evaluate the expression and significance of Nogo66 receptor (NgR) mRNA in adult ratsprime;optic nerve. Methods Optic and sciatic nerves of 8 adult rats were used to make the sections, which were divided into 3 groups: optic-nerve experimental group, sciatic-nerve control group, and optic-nerve negative control group. In situ hybridization was used to observe the expression of NgR mRNA in optic nerve and sciatic nerve. Results The expression of NgR mRNA in the 8 rats was positive in optic nerve and negative in sciatic nerve. The positive signals were arranged along the long axis of optic nerve. Conclusion The expression of NgR mRNA is positive in optic nerve while negative in sciatic nerve in adult rats, which suggests that the positive expression and distribution of NgR may be related to the poor regenerate ability of optic nerves. (Chin J Ocul Fundus Dis, 2005,21:246-248)

  • Effect of basic fibroblast growth factor on expression of apoptosisrelated genes in retinal ischemiareperfusion injury

    ObjectiveTo investigate the effect of basic fibroblast growth factor (bFGF) on expression of apoptosisrelated genes in retinal ischemiareperfusion injury (RIRI).MethodsTwentyeight rats were divided into normal, ischemia and treatment group randomly; and the latter two groups were subdivided into 6 subgroups according to different time points: 1 hour, 6, 12, 24, 48, and 72 hours after reperfusion. The rats′ model of experimental RIRI was established. After intravitreously injected with bFGF (treatment group) or balanced saline solution (ischemia group), the expressions of wide type p53 (WTp53),c-fos, and c-jun in each subgroups were detected by streptavidinbiotin complex of immunohistochemistry.ResultIn ischemia group, the expression of WTp53,c-fos and c-jun was found 6 hours after reperfusion, reached the peak at the 24th hour after reperfusion, kept expressing bly at the 48th hour, and decreased obviously at the 72nd hour. In treatment group, the rule of changes of expression of WTp53, c-fos and c-jun was similar to which in ischemia group, except that the expression amount was obvious decreased. There was statistical significance of the expression of WTp53, c-fos and c-jun between the ischemia and treatment group 6-48 hours after reperfusion (P<0.05). ConclusionThe expression of WTp53,c-fos,and c-jun in retinal ganglion cell layer and inner nuclear layer may increase led by RIRI;WTp53,c-fos,and c-jun may be involved in the generant mechanisms of RIRI by playing parts in apoptosis;bFGF can inhibit the increase of expression of WTp53,c-fos,and c-jun in RIRI.Thus, which may has therapeutic effect on RIRI.( Chin J Ocul Fundus Dis,2005,21:310-313)

  • Protein expression of genes related to apoptosis in retina of human fetus

    Purpose To study the proteins expression of genes related to apoptosis of retinal cells in development of human fetus. Methods Fifty cases of retinas of human fetus aged from 12 to 38 we eks were collected and paraffin embedded sections were made. Immunohistochemical method was used. Results Fas protein was expressed by cells of ganglion cell layer, inner and outer nuclear later, which were just formed on 16th week. It was not expressed until 38th week, Fas(+) staining appeared in layers of retina. Fas-L(+) staining was detected in cells of layers of retina on 26th week and the positive staining located in ganglion cell layer on 32th week. Neuronal fiber layer was Fas-L positive. Bax positive staini ng was detected on 8th week. Bax positive nucleus were observed mainly in GCL and ONL on 16th week. It was in INL on 24th week and in Muuml;ller cells inner terminates on 26th week. After this time, all cells of retina were bax immune ne gative staining. Bcl-2(+) staining appeared in differentiating neuroblastic layer on 16th week. Beginning on 24th week, bcl-2 (+) staining was observed in glial cells of GCL and inner terminates of Muuml;ller cell. Conclusion Apoptosis of developing retinal cell may be Fas/Fas-L independent and bax may be involved in apoptosis of the cells. (Chin J Ocul Fundus Dis, 2001,17:55-57)

  • VEGF mRNA expression in retinal neovascular tissue in rabbit eyes with retinal vein occlusion 

    Objective To evaluate the possible role of vascular endothelial growth factor (VEGF) in the pathogenesis of retinal neovascularization resulting from retinal ischemia. Methods Retinal vein occlusion of rabbits model was established with argon laser photo coagulation to retinal vein.The VEGF mRNA expression was observed in ischemic retina and retinal neovascular tissue with in situ hybridization technique. Results VEGF mRNA expression was detected in inner ischemic retina and retinal neovascular tissue that were corespondent to the distribution of retinal ischemia.The best VEGF mRNA expression was detected in retinal neovascular tissue. Conclusion VEGF might play an important role in the pathogenesis of vascular proliferative retinopathy. (Chin J Ocul Fundus Dis, 2001,17:5-7)
