  • Effect of Insulin Interventional Therapy on Hindlimb Ischemia of Rats with Diabetic

ObjectiveTo study the effect of exogenous insulin on inducing angiogenesis and the expression of vascular endothelial growth factor (VEGF) of hindlimb ischemia of rats with diabetic. MethodsThe hindlimb ischemic model of diabetic rat was established by the ligation of femoral blood vessels of hindlimb in twenty healthy male SD rats and which were divided into model group (n=10) and treatment group (n=10). Another 10 normal rats were selected as control group. Then the expression of VEGF protein and capillary density of muscle tissues of rat hindlimb were detected by Western blot analysis and alkaline phosphatase (APK) stain method, respectively. ResultsThe differences of body weight and blood glucose level of rats before operation and on day 7 after operation were not significant in the control group (Pgt;0.05). The body weight of rat was significantly lower on day 7 after operation than that before operation in the model group (Plt;0.05), while the difference of blood glucose level of rats was not significant in the model group (Pgt;0.05). The body weight and blood glucose level of rat significantly decreased in the treatment group on day 7 after subcutaneous injection of insulin as compared with the level before operation (Plt;0.05). Compared with the control group, the body weight decreased and blood glucose level increased in the model group and treatment group, and the difference was significant (Plt;0.05, Plt;0.01). The body weight of rat in the treatment group was not different from that in the model group (Pgt;0.05), but the blood glucose level of rat on day 7 after operation in the treatment group was significantly lower than that in the model group (Plt;0.05). The relative expression of VEGF protein of muscle tissues of ischemic hindlimbs in the treatment group (155.06±10.26) was significantly higher than that in the model group (94.30±11.23), Plt;0.05, while no expression was found in the control group. The capillary density of muscle tissues of right hindlimb in the control group was significantly higher than that in the model group or treatment group (Plt;0.05), and furthermore, which was higher in the treatment group than that in the model group (Plt;0.05). The difference of capillary density of muscle tissues of left hindlimb was not different among three groups (Pgt;0.05). The capillary density of muscle tissues of right hindlimb was not different from that of left hindlimb in the control group (Pgt;0.05) and which was significantly lower than that of left hindlimb in the model group or treatment group (Plt;0.05). ConclusionInsulin may increase the expression of VEGF protein in ischemic muscle tissue of diabetic rats and protect the ischemic muscle.

  • Efficacy Evaluation of Argatroban in Treatment of Lower Extremity Deep Vein Thrombosis

  • Correlation Between Homocysteine and AntiPhospholipid Antibodies and the Formation and Recurrence of Thrombus in Deep Vein.

【Abstract】ObjectiveTo detect the levels of homocysteine (Hcy) and anti-phospholipid antibodies (APLA) in the hematoplasma of the patients with deep venous thrombosis (DVT), discuss the reason of DVT recurrence and search for the predictors of it. MethodsSixty cases with DVT in our department from January 2001 to April 2003 were collected, which were divided equally into two groups as primary and recurrent, and first degree relative of the 30 DVT recurrent patients were also collected. The author established a control group using 30 cases of outpatient clinic without varicose veins of lower extremity or insufficient venae profundae. Hcy was detected with fluorescence polarization immunoassay (FPIA) and APLA 〔anticardiolipin antibody, ACLA (IgG, IgM); lupus antibody (LA)〕were detected with enzyme linked immunosorbent assay (ELISA). Odds ratios (OR) were also calculated to assess the relative risk of each study group. ResultsThe values of Hcy and ACLA (IgG, IgM) in the primary group and recurrent group were both significantly higher than those of control group and first degree relative group of DVT recurrent patients (Plt;0.01). The values of Hcy in first degree relative group of DVT recurrent patients was were also much higher than that of control group (Plt;0.05). The 90% quantity of Hcy in control group was 13.1 μmol/L and if taking it as the normal value, the number of cases exceeded this value in primary group, recurrent group and first degree relative group of DVT recurrent patients were 14, 21 and 13 and the OR of them were 2.31, 2.20 and 1.90, respectively. The positive rates of LA were not statistically significant in each group. Conclusion The values of Hcy and APLA both rise in the hematoplasma of DVT patients. There are close relationship between the levels of Hcy and APLA and DVT. The recurrence of DVT is related to high level of Hcy and Hcy is possibly a predictor of DVT.

  • Analysis of Cause of Death in Recipient Rats after Modified Orthotopic Liver Transplantations and the Countermeasure

【Abstract】ObjectiveTo investigate the causes of death in recipient rats after modified orthotopic liver transplantations and the countermeasure.MethodsTwo hundred and ten orthotopic liver transplantations were performed in Wistar and SD rats weighing 250~300 g through modified Kamada’s cuff technique, and to investigate the causes of death and preventive measures. ResultsIt took about (41±5) min, (13±2) min and (45±5) min for donor operation, donated liver trimming and recipient operation respectively. The anhepatic phase was (20±5) min. The 4week survival after transplantation was 90.5%. Nineteen rats died during and after transplantation. The causes of death included: bleeding of suprahepatic inferior vena cava (n=5 ), infrahepatic vena cava thrombosis (n=2), biliary obstruction (n=3 ), portal vein constriction and thrombosis (n=2 ), liver injury(n=1), bleeding of left subphrenic vein (n=2 ), infection (n=2 ), excessively deep anesthesia (n=1), and respiratory failure (n=1). ConclusionThe sophisticated microsurgical technique and the delicate surgical manipulation are very important in preventing operative complications, and the short anhepatic phase of recipient is the key to animal survival, and proper anesthesia is also important to the success of operations. Improving the condition of operation and reasonable use of antibiotics are necessary in preventing infections.
