  • Intra-pulmonary Artery Infusion of Antitumor Necrosis Factor-α Antibody Attenuates Lung Injury after Cardiopu-lmonary Bypass

    Objective To investigate the protective effects of antitumor necrosis factor-α antibody (TNF-αAb) on lung injury after cardiopulmonary bypass (CPB) and their mechanisms. Methods Forty healthy New Zealand white rabbits,weighting 2.0-2.5 kg,male or female,were randomly divided into 4 groups with 10 rabbits in each group. In groupⅠ,the rabbits received CPB and pulmonary arterial perfusion. In group Ⅱ,the rabbits received CPB and pulmonary arterial perfusion with TNF-αAb. In group Ⅲ,the rabbits received CPB only. In group Ⅳ,the rabbits only received sham surgery. Neutrophils count,TNF-α and malondialdehyde (MDA) concentrations of the blood samples from the left and right atrium as well as oxygenation index were examined before and after CPB in the 4 groups. Pathological and ultrastructural changes of the lung tissues were observed under light and electron microscopes. Lung water content,TNF-α mRNA and apoptoticindex of the lung tissues were measured at different time points. Results Compared with group Ⅳ,after CPB,the rabbitsin group Ⅰ to group Ⅲ showed significantly higher blood levels of neutrophils count,TNF-α and MDA(P<0.05),higherTNF-α mRNA expression,apoptosis index and water content of the lung tissues (P<0.05),and significantly lower oxyg-enation index (P<0.05) as well as considerable pathomorphological changes in the lung tissues. Compared with group Ⅱ,after CPB,the rabbits in groups Ⅰ and Ⅲ had significantly higher blood concentrations of TNF-α (5 minutes after aortic declamping,220.43±16.44 pg/ml vs.185.27±11.78 pg/ml,P<0.05;249.99±14.09 pg/ml vs.185.27±11.78 pg/ml,P<0.05),significantly higher apoptosis index (at the time of CPB termination,60.7‰±13.09‰ vs. 37.9‰±7.78‰,P<0.05;59.6‰±7.74‰ vs. 37.9‰±7.78‰,P<0.05),significantly higher blood levels of neutrophils count and MDA (P<0.05),significantly higher TNF-α mRNA expression and water content of the lung tissues (P<0.05),and significantly loweroxygenation index (P<0.05) as well as considerable pathomorphological changes in the lung tissues. Compared with groupⅠ,rabbits in group Ⅲ had significantly higher above parameters (P<0.05) but lower oxygenation index (P<0.05) only at 30 minutes after the start of CPB. Conclusion Pulmonary artery perfusion with TNF-αAb can significantly attenuate inflammatory lung injury and apoptosis of the lung tissues during CPB.

  • DIFFERENCE IN SENSITIVITIES OF CONJUNCTIVA FIBROBLASTS OF HUMAN EMBRYO AND RABBIT TO ANTINEOPLASTICS

    We compared the sensitivities of human embryo conjtmctival fihroblasts(HECF)and rabbit conjunctival fibroblasts(RCF)m five anlineoplasties in vitro.When the concentration of vincrismum and doxorubicin was 0.001~10mg/L.5-FU was 1~1000mg/L and cisplatin was 0.01~10mg/L,the sensitivities of HECF to tile drugs were lower than that of RCF (Plt;0.01).while the difference of the sensitivilics be;ween HECF and RCF to VP-16 was not significant (P<0.05). The results suggested that the selection of therapeutic agents for intraocular proliferative disease wilh'hunmn conjunctiva fihroblasts may be more valuable than that with RCF.   • Investigation on the Current Protection Situation for Hematology Department Nurses against Antitumor Drugs Exposure during Chemotherapy

    目的 了解血液科护士在对患者抗肿瘤药物治疗护理过程中的防护现状。 方法 2009年12月-2010年3月对血液科一、二病区和骨髓移植层流病房的护理人员,采用便利抽样方式进行问卷调查。 结果 血液科化学疗法防护现状与国际水平有差距,护理人员虽有一定的防护意识,但仍较薄弱。 结论 加强护理人员化学治疗药物毒性及防护知识的教育,提高防范意识,并制定严格的防护措施及管理制度是确保护理人员职业安全的根本与途径。

  • Analysis of Use of Antineoplastics in 17 Hospitals of Chengdu Area from 2007 to 2008

    【摘要】 目的 了解2007年-2008年成都地区17家医院抗肿瘤药物的使用情况。 方法 以销售金额、用药频度(DDDs)对2007年、2008年成都地区17家医院抗肿瘤药物消耗数据进行分类统计、综合分析、对比。 结果 2008年成都地区17家医院抗肿瘤药物用药金额增长幅度较大,各亚类抗肿瘤药物中其它类抗肿瘤药物类、植物来源的抗肿瘤药、抗代谢药金额排序依次占前3位。销售金额前100位药物中,肿瘤药物占据8位,大多数是其它类抗肿瘤药物,且其用药频度也靠前。 结论 抗肿瘤药物用药金额和用药量都将快速增长,其中其它类抗肿瘤药物和植物来源的抗肿瘤药的发展尤被看好。【Abstract】 Objective To evaluate the situation of antineoplastics used in Chengdu area from 2007 to 2008. Methods The consumption data of antineoplastics used in 17 hospitals of Chengdu area in 2007 and 2008 were classified, and the consumption sum and defined daily doses (DDDs) were analyzed. Results The total consumption sum of antineoplastics in 2008 increased sharply. Other antineoplastics, herbal-sourced antineoplastics and antimetabolism drugs ranked in the first three places. Among the top drugs in terms of consumption sum, antineoplastics took 8 places. Conclusion The consumption sum and DDDs of antineoplastics, especially other antineoplastics and herbal-sourced antineoplastics, will increase rapidly.

  • Curative Effect of Homoharringtonine United Program on Patients with Chronic Myelocytic Leukemia

    【摘要】 目的 探讨高三尖杉酯碱(HHT)联合三氧化二砷(As2O3)、羟基脲治疗高白细胞慢性粒细胞白血病(CML)的疗效和毒副作用。 方法 对2004年11月-2009年12月行高三尖杉酯碱联合As2O3、羟基脲治疗高白细胞CML患者21例的疗效和不良反应进行分析。 结果 经高三尖杉酯碱联合As2O3、羟基脲治疗后3~6个疗程,21例CML患者中完全缓解15例,占71%;部分缓解2例,占10%;总有效率81%;未缓解4例,占19%。不良反应仅表现乏力、纳差、恶心、脱发。 结论 HHT联合As2O3、羟基脲是治疗高白细胞CML一种有效率高,临床容易开展,安全性好的方法。【Abstract】 Objective To investigate the effects and side effects of homoharringtonine combined with arsenic trioxide or hydroxycarbamide on hyperleukocytic chronic myelocytic leukemia (CML). Methods The therapeutic effects and adverse effects of homoharringtonine combined with arsenic trioxide or hydroxycarbamide administered in 21 cases with CML from November 2004 to December 2009. Results In 21 patients with CML, the CR rate was 71% (15/21) and PR rate was 10% (2/21). The total respondence rate was 81% and the side effects were slight. The side effect only shows short of strength, appetite decrease, naupathia, hair loss and no heart side effect. Conclusion Homoharringtonine combined with arsenic trioxide or hydroxycabamide has a high effect rate on CML with good safety.

  • 5-FU Loaded Nanofibers Membrane for Controlled Release Effect Against Colon Cancer Cell in Vitro

    Objective To investigate the controlled release effect and the anti-cancer cell ability of a 5-FU loaded poly-L-lactic acid (PLLA) nanofibers membrane blending with keratin. Methods Making PLLA and keratin mix together and crosslinking to generate blending solution. Then the anti-cancer drug 5-FU was added into the solution to fabricate nanofibers membrane by high voltage electrospinning method. The micro morphology was observed by scanning electron microscope (SEM). The controlled release effect of 5-FU from the nanofibers membrane was measured by high performance liquid chromatography (HPLC). The cytotoxicity of 5-FU/PLLA keratin nanofibers membrane was evaluated by using 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay on HCT116 cell lines. At the meantime, cell growth morphology of HCT116 in experiment group were observed by microscope and transmission electron microscope. Results 5-FU could be dispersed homogeneous in the PLLA/keratin nanofibers membrane through SEM. HPLC suggested that 5-FU could be diffused out from the fibers slowly and uniformly, which corresponded the zero order kinetics basically. After different treatment, the longer time the 5-FU/PLLA keratin nanofibers (experiment group) immerse in the medium, the much more swelling, apoptosis, and necrocytosis of the cells were observed. The cell viability for experiment group was (47.5±2.8)% by MTT, while the PLLA keratin nanofibers without 5-FU had no significant impact on cell viability (93.9±2.8)%, which was statistic significance (P<0.01). Conclusion 5-FU/PLLA keratin nanofibers membrane owns good controlled release effect and satisfies cell inhibitory effect against HCT116 cells in vitro,which suggested that it has a promising prospect for clinical therapy.

  • EFFECTS OF SPLENECTOMY ON ANTI-TUMOR IMMUNITY DURING THE INDUCTION OF HEPATOCELLULAR CARCINOMA IN RATS

    Objective To study the effect of splenectomy on the anti-tumor immunity in rats with induced hepatocellular carcinoma (HCC). Methods At the second and fourth month of the induced HCC, the NK cell activity, TNF-α level and total lymphcyte in blood were measured in the group of splenectomy and the control group. Results There were no different in the total lymphcyte and TNF-α in the blood in two groups, but there were significant difference in the NK cell activity between the group of splenectomy and the control group (P<0.05). Conclusion There are some change in the anti-tumor immunity after splenectomy in rats, in which NK cell activity is at low level continuously. TNF-α isn′t affected after the second month after splenectomy.

  • 褪黑素对子宫内膜癌的影响

    子宫内膜癌是女性生殖道三大恶性肿瘤之一,病因迄今尚不明确,可能的危险因素有:肥胖、高血压、糖尿病、外源性雌激素的增加、高雌激素导致的初潮提前和绝经推后、卵巢肿瘤等。褪黑素作为一种主要由松果体分泌的神经内分泌激素,有调节睡眠/觉醒周期、免疫调节、细胞凋亡调节及抗氧化等多种生理功能。由于其抗氧化、免疫刺激、诱导凋亡等特性,在许多肿瘤的抑制和治疗上有一定效果。同时还具有调节血脂、血压、血糖,调节卵巢功能,抑制子宫内膜增生和子宫内膜癌细胞增殖,恢复子宫内膜等作用。因此,褪黑素水平的降低会增加子宫内膜癌的发病风险,且对其治疗有一定作用。现就褪黑素对子宫内膜癌的影响作一综述。
