【Abstract】Objective To investigate the significance of cyclin D1 and p53 protein expression in synchronous breast carcinoma and fibrosarcoma in rats. Methods Immunohistochemical SP methods was used to study the expression of cyclin D1 and p53 in synchronous breast carcinoma and fibrosarcoma induced by DMBA in rats.Results There was no expression of cyclin D1 and p53 in normal breast tissue. In atypical hyperplasia of mammary, there was overexpression of cyclin D1(7/14) and no expression of p53. The overexpression of cyclin D1 and p53 were detected in breast carcinoma (8/18,7/18 respectively) and fibrosarcoma (9/14,5/14 respectively). There was no expression of cyclin D1 and p53 in adjacent sarcoma.The expression of cyclin D1 and p53 protein was associated with histological grading, and showed inverse relation between them. Conclusion There are cyclin D1 and p53 protein overexpression in the synchronous breast carcinoma and fibrosarcoma induced by DMBA in rats. Cyclin D1 may paticipate in the course of the carcinogenesis of breast carcinoma and fibrosarcoma in rats, and p53 protein overexpression may relate to the degree of malignancy of the tumors.
Objective To reveal the association between the single nucleotide polymorphism (SNP) of v-maf musculoaponeurotic fibrosarcoma oncogene homolog B (MAFB) gene rs17820943 locus and non-syndromic cleft l ip with or without cleft palate (NSCL/P) in the southern Chinese Han population. Methods Genotyping of MAFB gene rs17820943 polymorphism was carried out in 300 patients with NSCL/P, 354 normal controls, and an additional 168 case-parent trios with matrix-assisted laser desorption/ionisation time-of-fl ight (MALDI-TOF) mass spectrometry. Then based on the genotypingresults, both a case-control association study and a case-parent trio association study were performed. Results Significant differences were found in the allele and genotype frequencies of rs17820943 locus between case and control groups (Pallele=0.001 and Pgenotype=0.002, respectively). To be specific, the odds radio (OR) values and 95% confidence interval (95%CI) of allele T (frequencies of cases ∶ controls = 0.358 ∶ 0.448) and genotype TT (frequencies of cases ∶ controls = 0.110 ∶ 0.195) were ORT = 0.69 (95%CI: 0.55-0.86) and ORTT = 0.43 (95%CI: 0.26-0.70), respectively. Subsequent case-parent trio analysis also indicated an association between MAFB rs17820943 variant and the risk of NSCL/P (ORT vs. C = 0.55, 95%CI: 0.41-0.75, P value of transmission disequilibrium test was 0.000). Conclusion Polymorphism of MAFB gene rs17820943 locus is associated with NSCL/P in the southern Chinese Han population; MAFB rs17820943 variant may be a susceptible gene of NSCL/P.
Objective To investigate the method and effectiveness of wide local excision combined with Mohs micrographic surgery for dermatofibrosarcoma protuberans (DFSP). Methods Between January 2007 and January 2010, 17 patients with DFSP were treated. There were 9 males and 8 females with an average age of 33.2 years (range, 16-55 years). Thelesions were located at head and neck (2 cases), trunk (12 cases), extremity (2 cases), and perineal region (1 case). There were 6 cases of primary DFSP and 11 cases of relapsed DFSP. The lesions presented as single or multitude nodules or fusion nodules with skin withering, scar, en plaque in the center and with ill-defined margins. The diameter of lesions ranged from 0.8 to 9.7 cm (mean, 4.3 cm). No distant metastasis or lymphatic metastasis occurred in all cases. After tumors resection by wide local excision combined with Mohs micrographic surgery, the wounds were repaired by direct suture in 3 patients, skin grafting in 9 patients, and local skin flap in 5 patients. Results Wide local excision and Mohs micrographic surgery were carried out once in 13 patients, twice in 3 patients, and three times in 1 patient with an average operation time of 98.6 minutes (range, 56-219 minutes). Primary heal ing of wound and donor site were achieved with no necrosis of skin grafting and skin flap. All patients were followed up 8-34 months (mean, 21.7 months) with no recurrence. Conclusion Wide local excision combined with Mohs micrographic surgery could treat DFSP, which has the advantages of shorter operation time, radical resection, and less injury.
目的 探讨硬化性上皮样纤维肉瘤(SEF)的临床病理学特征。 方法 对2012年3月收治的l例SEF患者进行组织学观察、免疫组织化学检查并复习相关文献。 结果 SEF主要以细胞质透亮的上皮样肿瘤细胞伴大量透明变性胶原纤维为特征,瘤细胞排列主要呈条索状、巢状。肿瘤细胞波形蛋白阳性表达,细胞角蛋白和平滑肌肌动蛋白阴性表达。 结论 SEF是一种较为罕见的软组织肿瘤,应注意与一些具有上皮样形态和硬化性间质的肿瘤相鉴别。
