  • The reporting quality of intervention animal studies published in Chinese journals: a quantitative analysis

    ObjectivesUsing the ARRIVE guidelines (Animal Research Reporting: In Vivo Experiments Guidelines) to carry out a retrospective study of the reporting quality of animal studies published in Chinese journals.MethodsWe searched databases including CNKI, WanFang Data, VIP and CBM to July, 2018. Four reviewers independently screened literatures and extracted data. The ARRIVE guidelines were used to assess reporting quality and the comparative analysis based on different published time.ResultsA total of 4 342 studies were included. About the cited frequency, 73.03% studies were ≤5, and merely 29.04% studies were published in journals of CSCD. The assessment results showed that the number of reported items with "low risk" in the ARRIVE guidelines, which have 20 items, that meaning 39 sub items, more than half of sub items (51.28%, 20/39) rated as "low risk" had a compliance rate of less than 50%. Among them, 65.00% (13/20) of sub items had a lower rate of compliance with "low risk" than 10%.ConclusionThe reporting quality of domestic animal studies is generally low. The coincidence rate of domestic animal studies has been improved to some extent in most of items after the ARRIVE guidelines published, however, some items of methodology, results and conclusions had problems with insufficient reporting. Therefore, we suggest that it is necessary to popularize the ARRIVE guidelines, advocate more researchers following the ARRIVE guidelines and promote endorsement of the ARRIVE Guideline by Chinese Journals to improve the design, implementation and reporting of animal experiments, and ultimately enhance the quality of animal studies.

  • A Survey on Awareness of the ARRIVE Guideline and GSPC in Researchers Field in Animal Experiments Field in Lanzhou City

    ObjectiveTo investigate the awareness situation on the ARRIVE guideline and the Gold Standard Publication Checklist (GSPC) of animal experiments in researchers in animal experiments field in Lanzhou city, in order to improve the promotion of the two reporting guidelines in China. MethodsA self-designed questionnaire was used to investigate the clinical graduate students and teachers in medical college in Lanzhou city. The investigation contents mainly included the basic information of the respondents, the awareness situation on the ARRIVE guideline, GSPC and other medical reporting guidelines. SPSS 21.0 software was used for data analysis. ResultsA total of 329 questionnaires (40 were from teachers and 289 were from graduate students) were issued, of which, 287 questionnaires were effective. The results showed that the awareness rate on the ARRIVE guideline and GSPC in clinical graduate students and teachers in medical college in Lanzhou city were 11.8% and 12.5%, respectively, and there was no significant difference between students and teachers in awareness rate (P=0.903). The survey approaches, the age, education, job, and the organization of the respondents were all not the influence factors of awareness rate (P>0.05). The respondents knew about the reporting guidelines mainly through the website (33.4%), related studies (21.2%) and academic reports (17.4%). ConclusionThe awareness rate on the ARRIVE guideline and GSPC is relative low in researchers in animal experiments field in Lanzhou city, and it needed to take purposeful measures to promote and popularize them.

  • Endorsement of the ARRIVE Guideline and GSPC Checklist by Chinese Journals: A Survey of Journal Editors and Review of Journals'Instructions for Authors

    ObjectiveTo assess the endorsement of the ARRIVE guideline and the Gold Standard Publication Checklist (GSPC) by Chinese journals in animal experiments field and its incorporation into their editorial processes. MethodsChinese journals indexed by SCI, MEDLINE, CSCD or CSTPCD were included. The latest'instruction for authors' (IFA) of each included journals was downloaded and any text mentioning the ARRIVE guideline and GSPC was extracted. Subsequently, a self-designed questionnaire was used to investigate the editor of each included journals. The investigation contents mainly included the basic information of the respondents, the awareness situation on the ARRIVE guideline, GSPC and their incorporation into editorial and peer review processes. Results240 journals in animal experiments field from China were examined. A total of 240 questionnaires were issued, of which, 198 questionnaires were effective (response rate 82.5%). The results showed that all IFAs didn't mention the ARRIVE guideline or GSPC and the awareness rate on the ARRIVE guideline and GSPC in editors of Chinese journals was only 13.1%. Only 10.1% of the editors reported that they required authors to comply with the ARRIVE guideline and GSPC. And editors reported that they incorporated the two guidelines into their peer review (7.1%) and editorial processes (8.1%). ConclusionAt present, all Chinese journals'IFAs didn't mention the ARRIVE guideline or GSPC. The majority of editors surveyed are not familiar with the content of the ARRIVE guideline and GSPC. And it needs to take purposeful measures to promote and popularize them in order to improve the quality of animal experiment reports.

  • Stepwise method based on Wiener estimation for spectral reconstruction in spectroscopic Raman imaging

    Raman spectroscopy has demonstrated great potential in biomedical applications. However, spectroscopic Raman imaging is limited in the investigation of fast changing phenomena because of slow data acquisition. Our previous studies have indicated that spectroscopic Raman imaging can be significantly sped up using the approach of narrow-band imaging followed by spectral reconstruction. A multi-channel system was built to demonstrate the feasibility of fast wide-field spectroscopic Raman imaging using the approach of simultaneous narrow-band image acquisition followed by spectral reconstruction based on Wiener estimation in phantoms. To further improve the accuracy of reconstructed Raman spectra, we propose a stepwise spectral reconstruction method in this study, which can be combined with the earlier developed sequential weighted Wiener estimation to improve spectral reconstruction accuracy. The stepwise spectral reconstruction method first reconstructs the fluorescence background spectrum from narrow-band measurements and then the pure Raman narrow-band measurements can be estimated by subtracting the estimated fluorescence background from the overall narrow-band measurements. Thereafter, the pure Raman spectrum can be reconstructed from the estimated pure Raman narrow-band measurements. The result indicates that the stepwise spectral reconstruction method can improve spectral reconstruction accuracy significantly when combined with sequential weighted Wiener estimation, compared with the traditional Wiener estimation. In addition, qualitatively accurate cell Raman spectra were successfully reconstructed using the stepwise spectral reconstruction method from the narrow-band measurements acquired by a four-channel wide-field Raman spectroscopic imaging system. This method can potentially facilitate the adoption of spectroscopic Raman imaging to the investigation of fast changing phenomena. (C) 2017 Optical Society of America

  • Therapeutic Effects of Human Umbilical Cord-Derived Mesenchymal Stem Cells in Acute Lung Injury Mice

    The incidence and mortality of acute lung injury (ALI)/acute respiratory distress syndrome (ARDS) are still very high, but stem cells show some promise for its treatment. Here we found that intratracheal administration of human umbilical cord-mesenchymal stem cells (UC-MSCs) significantly improved survival and attenuated the lung inflammation in lipopolysaccharide (LPS)-induced ALI mice. We also used the proteins-chip and bioinformatics to analyze interactions between UC-MSCs treatment and immune-response alternations of ALI mice. Then we demonstrated that UC-MSCs could inhibit the inflammatory response of mouse macrophage in ALI mice, as well as enhance its IL-10 expression. We provide data to support the concept that the therapeutic capacity of UC-MSCs for ALI was primarily through paracrine secretion, particularly of prostaglandin-E2 (PGE2). Furthermore, we showed that UC-MSCs might secrete a panel of factors including GM-CSF, IL-6 and IL-13 to ameliorate ALI. Our study suggested that UC-MSCs could protect LPS-induced ALI model by immune regulation and paracrine factors, indicating that UC-MSCs should be a promising strategy for ALI/ARDS.

  • A Clinical Study of Spontaneous Pneumothorax in Elderly Patients

    目的:探讨老年人自发性气胸的临床特点、治疗及预后。方法:对本院在2005年11月至2008年4月间收治的79例老年人自发性气胸患者临床资料进行回顾性分析。结果:老年人自发性气胸大多有肺部基础疾病,临床表现缺乏特异性,本组误诊为慢性阻塞性肺病急性发作4例、左心衰2例、支气管哮喘1例。气胸类型: 张力性气胸47例(72.1%),闭合性气胸11例,交通性气胸21例。采用以肋间闭式引流的为主的治疗措施,效果好。结论:老年人自发性气胸大多有肺部基础疾病, 易误诊,气胸的类型以张力性气胸多见,治疗多需排气减压术,及早的排气减压可望缓解症状,缩短肺复张时间,减少患者住院天数, 降低死亡率,提高老年人生活质量。

  • Cordycepin induces autophagy-mediated c-FLIPL degradation and leads to apoptosis in human non-small cell lung cancer cells

    Cordycepin, a main active composition extracted from Cordyceps militaris, has been reported to exert anti-tumor activity in a broad spectrum of cancer types. However, the function of cordycepin on human non-small cell lung cancer cells is still obscure. Our present work showed that cordycepin inhibited cell growth by inducing apoptosis and autophagy in human NSCLC cells. Further study revealed that cordycepin triggered extrinsic apoptosis associated with down-regulation of c-FLIPL which suppresses the activity of caspase-8. And ectopic expression of c-FLIPL dramatically prevented cordycepin-caused apoptosis. Meanwhile, cordycepin stimulated autophagy through suppressing mTOR signaling pathway in lung cancer cells. When autophagy was blocked by Atg5 siRNA or PI3K inhibitor LY294002, the levels of apoptosis caused by cordycepin were obviously attenuated. In addition, suppression of autophagy could also elevate the level of c-FLIPL which indicated cordycepin-triggered autophagy promoted the degradation of c-FLIPL. Therefore, we conclude that cordycepin induces apoptosis through autophagy-mediated downregulation of c-FLIPL in human NSCLC cells. Taken together, our findings provide a novel prospect on the anti-tumor property of cordycepin, which may further prompt cordycepin to serve as a promising therapeutic approach in NSCLC treatment.

  • Non-toxic dose of liposomal honokiol suppresses metastasis of hepatocellular carcinoma through destabilizing EGFR and inhibiting the downstream pathways

    At present, there is no specific anti-metastasis drug in HCC treatment. Drugs used for primary HCC tumors and tumor metastasis are very similar, among which cytotoxic drugs are prevalent, such as cisplatin, doxorubicin and 5-FU. The EGFR pathway plays an important role in promoting hepatocellular carcinoma (HCC) metastasis. Hence, development of non-toxic anti-metastasis drugs, such as EGFR or downstream pathways inhibitors, is of great importance. In our present study, we found non-toxic dose of liposomal honokiol (LH) could inhibit the HCC metastasis by destabilizing EGFR and inhibiting the downstream pathways. Non-toxic dose of LH significantly inhibited the motility, migration and lamellipodia formation of HepG2 cells in vitro and decreased extravasation of HepG2 cells in a novel metastasis model of transgenic zebrafish. In two lung metastasis models (HepG2 and B16F10) and a spontaneous metastasis model of HepG2 cells, LH remarkably inhibited pulmonary metastasis and regional lymph nodes metastasis without obvious toxicity. Further study showed that destabilizing EGFR and inhibiting the downstream pathways were the main mechanisms of non-toxic dose of LH on metastasis inhibition. Our results provide the preclinical rationale and the underlying mechanisms of LH to suppress HCC metastasis, implicating LH as a potential therapeutic agent to block HCC metastasis without severe side effects.

  • Neuroprotective effects of BDNF and GDNF in intravitreally transplanted mesenchymal stem cells after optic nerve crush in mice

    AIM: To assess the neuro-protective effect of bone marrow mesenchymal stem cells (BMSCs) on retinal ganglion cells (RGCs) following optic nerve crush in mice. METHODS: C56BL/6J mice were treated with intravitreal injection of PBS, BMSCs, BDNF-interference BMSCs (BIM), and GDNF-interference BMSCs (GIM) following optic nerve crush, respectively. The number of surviving RGCs was determined by whole-mount retinas and frozen sections, while certain mRNA or protein was detected by q-PCR or ELISA, respectively. RESULTS: The density (cell number/mm(2)) of RGCs was 410.77 +/- 56.70 in the retina 21d after optic nerve crush without any treatment, compared to 1351.39 195.97 in the normal control (P<0.05). RGCs in BMSCs treated eyes was 625.07 +/- 89.64/mm(2), significantly higher than that of no or PBS treatment (P<0.05). While RGCs was even less in the retina with intravitreal injection of BIM (354.07 +/- 39.77) and GIM (326.67 +/- 33.37) than that without treatment (P<0.05). BMSCs injection improved the internal BDNF expression in retinas. CONCLUSION: Optic nerve crush caused rust loss of RGCs and intravitreally transplanted BMSCs at some extent protected RGCs from death. The effect of BMSCs and level of BDNF in retinas are both related to BDNF and GDNF expression in BMSCs.
