  • Experimental Study of Hemolysis in Vitro and Animal Testing for Fu Wai Axial Blood Pump

    Objective To provide a ventricular assist device for patients with heart failure, Fu Wai (FW) axial blood pump was developed for partly or totally to assist the left ventricular function. Vitro hemolysis and animals tests were also employed to test the hydromechanics and hemocompatibility of the FW left ventricular assist devices developed in Fu Wai hospital. Methods Using vitro test loop, FW axial blood pump has been used to evaluate the performance of hemolysis, the pump has also been tested for hemolysis characteristic through five sheep experiments. Results At 8 400 r/min, the pump generates 5 L/min flow against 100 mm Hg, the normalized index of hemolysis (NIH) was0.17±0.06 mg/L. The plasma free hemoglobin of in vivo tests was around 30 mg/dl. Conclusion The results obtained in vitro and in vivo testing indicate an acceptable design for the blood pump, further in vivo tests will be performed before clinical use.

  • Electrophysiological response in rabbits with normal and injured photoreceptor due to subretinal implantation of chip

    Objective To observe the changes of electrophysio logical results in rabbits with normal and injured photoreceptor due to subretinal implantation of chip. Methods Photoreceptor damage was induced by injection with NaIO3 solution in 22 out of 30 rabbits. A chip with the diameter of 3 mm made by the array composed of 90 microelectrodes photodiode and conjoint electrode was implanted into subretinal space or choroid of the right eyes of 22 rabbits with photoreceptor and 4 normal rabbits, and the left eyes were the control. The examinations of local flash-visual evoked potential (F-VEP), local flash-electroretinogram (F-ERG), full-field F-ERG and full-filed F-VEP were measured respectively.Another 4 rabbits underwent biocular extirpation for path ological examination . Results In 22 rabbits with photo-receptor damage, the amplitude of the main wave of local ERG was obviously higher in 11 eyes with chips than that in the control ones, and was also higher in 2 eyes with chips of the 4 mormal rabbits than that in the control eyes. No wave was found in an eye with retinal hole on the surface of the chip. The repeataility of main amplitude of local-VEP and full-field F-VEP is not satisfactory; no significant changes were observed between chip-implanted eyes and the control eyes examined by full-filed F-ERG. Conclusion The implanted chip may stimulate local retina and induce electrical activities after stimulated by light. (Chin J Ocul Fundus DIs, 2006, 22: 324-327)

  • The preliminary study on commitment differentiation of embryonic stem cells induced by the medium of cultured retinal neurons of SD rats

    Objective To investigate the possibility of commitment differentiation of embryonic stem cells induced by the medium of cultured retinal neurons of SD rats. Methods The medium from cultured retinal neurons of SD rats were collected, sterilized and mixed with DMEM medium according to 2∶3 proportion, ES cells were cultured with these mixed medium and were observed under the phase contrast microscope daily, the induced cells were identified by NF immunohistochemistry methods. Results The ES cells cultured with these mixed medium can differentiate into neuron-like structure, and the induced cells were positive in NF immunofluorescence staining. Conclusion The medium from cultured retinal neurons of SD rats can induce ES cells commitment differentiation into neuron-like structure. (Chin J Ocul Fundus Dis, 2002, 18: 134-136)

  • The relationship between electrophysiological and morphological properties of neurons in visual cortex of developing rat-intracellular staining study

    Objective To investigate the relationship between electrophysiological and morphological properties of neurons in visual cortex of developing rat, speculate the coincided degree between electrophysiological and morphological change and realize the mechanism of normal visual development. Methods Whole cell patch-clamp recording and intracellular staining were used to acquire cellular microelectrode recording in visual cortex from Sprague-Dawley rats (4~28 days old). The histological process was made. Results The differences of electrical feature between pyramidal cells and non-pyramidal cells were significant. The morphological maturity degree is different in developing visual cortex. Conclusion The different function of pyramidal and non-pyramidal cells in local integrition is reflected by their electrical feature in the process of visual development. In critical period of visual development, the coincision degree of the electrophysiological and morphological change in visual cortex is larger than that in the subcortex constructure. (Chin J Ocul Fundus Dis, 2001,17:289-292)

  • Induction and maintenance of immune deviation in different intraocular compartment

    Objective To determine whether the vitreous cavity and the subretinal space of different species animals support the induction of immune deviation to soluble antigen and to observe its maintenance time. Methods Bovine serum albumin(BSA)was used as a soluble antigen and inoculated into the anterior chamber(AC),the vitreous cavity(VC)and subretinal space of different animals(rat,rabbit and monkey)respectively.Recipient animals were immunized with BSA and complete Freundrsquo;sadjuvant. Delayed-type hypersensitivity(DTH)was assessed by skin challenge.The maintenance time of deviant immune response was evaluated in the fixed time interval. Results All the animals receiving intraocular antigen inoculation did not display DTH reaction.The maintenance time of immune deviation after inoculation of antigen in different sites is (1)anterior chamber:70 days in rat,90 days in rabbit,320 days in monkey;(2)vitreous cavity:100 days in rat,150 days in rabbit,360 days in monkey;(3)subretinal space:50 days in rat,70 days in rabbit,300 days in monkey. Conclusions The maintenance time of immune deviation varied with different species of animals and different intraocular compartment.The evolution of organisms is synchronous with that of immune system.
