  • COMPARATIVE STUDY OF EPIDERMAL GROWTH FACTOR AND BASIC FIBROBLAST GROWTH FACTOR ON WOUND HEALING

    OBJECTIVE The biological effects of recombinant human epidermal growth factor (rhEGF) and recombinant human fibroblast growth factor (rhFGF) were evaluated on the model of incised wounds in mini pigs. METHODS Total of 160 incised wounds in 16 mini pigs were divided into two groups (rhEGF group and rhFGF group), each containing 80 wounds. In rhEGF group, 60 incised wounds were treated with different dosages of rhEGF (50, 10 and 0.5 micrograms/wound), and another 20 wounds were treated with solvent as control group. In rhFGF group, all wounds were treated in the same way as described in rhEGF group, the dosages of rhFGF were 150, 90 and 30 U/cm2 respectively. The measurements of cavity volume and area in wound, histological examination were used to evaluate the results of wound healing. RESULTS The results showed that wound healing was accelerated in all wounds treated with rhEGF and rhFGF. In rhEGF group, the velocity of re-epithelialization was faster than that of rhFGF group, however, new granulation tissue in rhFGF was more than that of rhEGF group. CONCLUSION The results indicate that rhEGF and rhFGF can stimulate wound healing, however, the mechanisms and the biological effects involved in these processes are quite different. It suggests that it is better to use rhFGF in those wounds which need more granulation tissue formation and use rhEGF in the wounds which mainly need re-epithelialization.

  • EFFECTS OF BIOELECTRET COMPOSITES ON ANIMAL INCISED WOUND HEALING

    Abstract For the purpose of studying the effects on wound healing of three new bioelectret composites (BC) which are composed ofcollagen polyvinyl alcohol and epidermal growth factor (EGF), 30 rabbits were divided into 5 groups. Three wounds in round shape (1.8 cm in diameter) were made in each side of the back of the rabbits. The wounds of 1~3 groups were treated by one of the three BC respectively, group 4 treated by SDAg, and group 5 were treated by Normal saline as control. From observation of the growth of the granulation tissue, the reepithelization and the pathological assessment, it was shown that the quality of wound healing in all BC treated wounds was better than that in SDAg or the control. It indicated that the BC benefited the wound healing, and this might also be due to its bioelectric effects and the direct effects from growth factor.

  • EFFECT AND SIGNIFICANCE OF TETRANDRINE ON EPIDERMAL GROWTH FACTOR ANDITS RECEPTOR IN THE LUNG OF CONGENITAL DIAPHRAGMATIC HERNIARAT MODEL

    Objective To investigate the effect of the traditional Chinese medicine “Tetrandrine”(TET) and its significance on epidermal growth factor(EGF) and its receptor(EGFR) in the lung of nitrofen-induced congenital diaphragmatic hernia(CDH) rat model. Methods Twenty female rats were given maternal administration of a single oral dose (115 mg/rat) of nitrofen to induce CDH at 9.5 days after pregnancy and were dividedinto normal solution group(NS, n=5), dexamethasone group (Dex, n=5),tetrandrine group (TET, n=5) and Dex+TET group(n=5) at 18.5 days; 4 rats were given edible oil as controls. All fetuses were delivered by cesarean section at 21.5 days. Lung histologic evaluations and EGF, EGFR immunohistochemical staining and image analysis were performed. Results CDH was observed in 64 of the 137 rat fetuses (46.7%) in the experimental groups; no CHD was observed in 36 rat fetuses of control group. The lungs of CDH fetuses showed marked hypoplasia in NS group, in contrast to improved mesenchymal differentiationin that of Dex, TET, Dex+TET groups. The expression of EGF was weaker and weaker and that of EGFR was ber and ber as following order: NS, TET, DEX, T+D and control groups; showing significant differences between them (Plt;0.05). Conclusion Prenatal TET administration shows marked improvement in pulmonary hypoplasia through preregulating crest-time of EGF expression and upregulating EGFR expression in the lungs of nitrofen-induced CDH rat model. A combination of TET and Dex would generate evident synergistic effect.

  • THE EXPRESSION CHARACTERISTICS AND THEIR BIOLOGICAL SIGNIFICANCE OF EPIDERMAL GROWTH FACTOR AND BASIC FIBROBLAST GROWTH FACTOR IN RAT SKINS AT DIFFERENT DEVELOPMENT STAGES

    OBJECTIVE: To explore the localization and expression characteristics of epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) in rat skins at different development stages (embryonic, newborn, adult). METHODS: Skins from embryonic, newborn and adult rats were taken, and detected by immunohistochemical technique. RESULTS: Positive immunohistochemical signal of EGF could be found in skins from embryonic, newborn and adult rats, mainly in the cytoplasm of the epidermal cells, fibroblasts, hair follicle epithelial cells, and endothelial cells. With the increase in age, the expression amount of EGF was increased. The positive signal of bFGF could be found in skins of newborn and adult rats, while the signal of bFGF in skin of embryonic rats was negative. CONCLUSION: The results indicate that endogenous EGF plays important role in epidermal development in embryonic stage and wound healing in adult after injury. The negative expression of bFGF in skin of embryonic rat indicate that the absence of bFGF may be one of the reasons for the non-scar healing in embryonic stage.

  • Clinical Significance of Epidermal Growth Factor Receptor Mutations from Patients with Non-Small Cell Lung Cancer

    Objective To evaluate the clinical significance of epidermal growth factor receptor EGFR) mutations in the treatment of non-small cell lung cancer ( NSCLC) . Methods Plasma DNAs solated fromblood specimens of 170 NSCLC patients, who were admitted in the First Affiliated Hospital of uangzhou Medical College from December 2005 to December 2007, were subjected to the test of EGFR utant-enriched PCR. The correlation of mutant detection with clinical characteristics was analyzed as well.Results Out of the total 170 patients, EGFR mutations were identified in 77 cases ( 77 /170, 45. 3% ) .EGFR mutations were more frequent in the patients with adenocarcinoma ( P lt; 0. 001) and in the nonsmokers P =0. 001) . In the 33 patients treated with gefitinib, those with mutations ( + ) showed a higher esponse rate and prolonged progression-free survival after the treatment compared with those with mutations( - ) ( P =0. 001 and 0. 001, respectively) . Conclusions EGFR active mutations can be specifically and ensitively detected by EGFR mutant enriched PCR assay. Plasma EGFR mutants detection is valuable in uiding clinical decision.

  • PROLIFERATIVE EFFECT OF BASIC FIBROBLAST GROWTH FACTOR AND EPIDERMAL GROWTH FACTOR ON MUSCLE DERIVED STEM CELLS

    Objective To explore the effect of basic fibroblast growth factor(bFGF)and epidermal growth factor(EGF)on the growth of muscle derived stem cells(MDSCs). Methods MDSCs were isolated from hindlimb muscle of 15 new born Kunming mice through serial preplates. 2% fetal bovine serum-containing DMEM was used to induce MDSCs to differentiate into skeletal muscle lineage. The expressions of stem cell marker Sca-1 and skeletal musclecell marker αSarcomeric actin were examined by immunocytochemistry. The effect of bFGF and EGF on the proliferation of MDSCs was determined by MTT colorimetric microassay. The solo effect of bFGF or EGF at different concentrations (6.25,12.50, 25.00, 50.00, and 100.00 ng/ml) was examined at 96 h and the combined effect (100.00 ng/ml) was examined at 24,48,72 and 96 h.Results MDSCs were successfully isolated from the hindlimb of neonatal mice. Over 90% of MDSCs showed Sca-1 positive immunoreactivity. MDSCs could give rise to α Sarcomeric actin positive myotubes in differentiation cultures. The proliferative effect of bFGF and EGF on MDSCs increased with the elevated concentration.bFGF began to show significant proliferative effect at 12.50 ng/ml (P<0.05). The effect increased significantly when the concentration reached 25.00 ng/ml from 12.50 ng/ml (P<0.01) and reached a saturation point. The effect at 50.00 ng/ml or 100.00 ng/ml showed no significant increase when compared with thatat 25.00 ng/ml. EGF had a similar effect to bFGF except that the saturation concentration was 50.00 ng/ml. EGF showed significant effect at 72 h and bFGF at 96 h (Plt;0.01). When they were applied together, significant effect was shownat 24 h (Plt;0.01) and much higher effect was observed at 48, 72 and 96 h (Plt;0.05). Conclusion Both bFGF and EGF can promote the proliferation of MDSCs. The combined application reacts faster and ber.

  • Expression and Clinical Significance of Phosphatase and Tensin Homologue Deleted on Chromosome Ten,Epidermal Growth Factor Receptor and Ki67 in Human Thymic Tumors

    Abstract: Objective To investigate the expression and correlation of phosphatase and tensin homologue deleted on chromosome ten(PTEN), epidermal growth factor receptor(EGFR) and Ki-67 in human thymic tumors, and their possible role in tumor genesis, infiltration and metastasis. Methods The expression of PTEN, EGFR and Ki-67 were detected by using SP immunohistochemical technique in 45 cases of thymic tumors and 5 cases of normal thymic tissues. Results In 5 cases of normal thymic tissues, the expression of PTEN was bly positive, whereas EGFR and Ki -67 were weakly positive or negative. In 45 cases of thymic tumors, the positive ratio of PTEN were significantly reduced from benign thymoma, invasive thymoma to thymic carcinoma (χ2=7.808, P=0.020), but the positive ratio of EGFR and Ki-67 were gradually increased(χ2=8.032, 0.018,7.006;P=0.030). The positive ratio of PTEN, EGFR and Ki-67 protein were significantly related to Levine classification, Masaoka staging and lymph node metastasis (Plt;0.05). PTEN positive cases were negatively correlated with EGFR and Ki-67(r=-0.632,-0.653;Plt;0.01), EGFR positive cases were positively correlated with Ki-67 in thymic tumors(r=0.807,Plt;0.01). Conclusions Reduced or absent PTEN and increased EGFR and Ki-67 expression might play an important role in the genesis, invasiveness and metastasis of thymic tumors. The expression of PTEN is bly associated with the expression of abnormal EGFR and Ki-67. Detection of the three protein expressions simultaneously might be more helpful in making an early diagnosis of the tumors jndgement of theirs malignant degree,invasiveness and metastasis capacity, as well as the prognosis.

  • Growth factor effects on the proliferation of human retinal glial cell in culture

    Objective:To study the effects of growth factor on the proliferation of the cultured huamn retinal glial cells. Methods:EGF(0.5~100.0ng/ml) and NGF (0.5~10.0ng/ml) were added to cultures of human retinal glial cells and the proliferation rates of the cells were measured by MTT method. Results:EGF at a dosage ranging from 0.5ng/ml to 100.0ng/ml and NGF (0.05~10.0ng/ml) stimulated the cellular proliferation effectively with their EC 50 of 17ng/ml and 0.7 ng/ml respectively. Conclusion:Both EGF and NGF NGF had an effective stimulation on human retinal glial cell proliferation.They may play a role in the formation of PVR. (Chin J Ocul Fundus Dis,1998,14:33-34)

  • EXPRESSION OF EPIDERMAL GROWTH FACTOR RECEPTOR IN HUMAN THYROID CARCINOMAS

    To investigate the significance of epidermal growth factor receptor (EGFR) in thyroid carcinoma, the expression of EGFR in 81 samples of thyroid carcinoma were determined by immunohistochemical SP method and comparison among thyroid carcinoma, thyroid adenomas and normal thyroid tissue adjacent to the cancer were made. The results showed: EGFR expression was positive in 45 cases (55.6%) of thyroid carcinoma with no positive expression either in thyroid adenomas or normal thyroid tissue adjacent to the cancer (P<0.01). There were no statistical differences between EGFR positive rate and thyroid carcinomatous pathological type, clinical stage, depth of invasion, lymph node metastasis or patients′ postoperative survival time (P>0.05). This data suggests that expression of EGFR in thyroid carcinoma is associated with its autonomous growth and malignant phenotype, but it is probably not a useful index for assessing the biological behavious and prognosis of thyroid carcinoma.

  • EGF and bFGF effects on DNA synthesis in human retinal pigment epithelial cells

    Porpose To investigate the optimal concentration of epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) on DNA synthesis and their synergism indensity arrested human retinal pigment epithelial (RPE) cells. Methods Growth factor effects in cultured human RPE of the 6th generation were assessed by [3 H]-thymidine incorporation and radioautography. Results EGF and bFGF were potent stimulators when used alone,and their optimal concentrations were 10ng/ml in DMEM and 1ng/ml in 2% serum DMEM.When used in combination (10ng/ml EGF and 10ng/ml bFGF),they caused a significant enhancement of [3 H]-thymidine incorporation about 2.96 times. Conclusion EGF and bFGF were potent stimulators in RPE cells,and demonstrated synergism in their action. (Chin J Ocul Fundus Dis,1998,14:98-100)
