  • Value of MUC1 and MUC3 in the Calcareous Cholocystitis and Cholecystic Adenomatoid Polyps

    bjective To study the change of mucins of expression in lithic cholecystitis and cholecystic adenomatiod polyps. MethodsMUC1 and MUC3 were detected in the mucosa of human normal gallbladders (20 cases, control group), of calcareous cholecystitis (38 cases, calcareous group) and of adenomatoid polyps (18 cases, polyp group) with immunohistochemical stains and Western blotting methods. ResultsThe positive rate and optical density values of MUC1 were increased significantly in calcareous and polyp group vs control group (P<0.01), otherwise, MUC3 was decreased markedly (P<0.01). Conclusion The expressions of MUC1, MUC3 were not synchronization in different lesions of cholecyst.

  • ROS/Src /JNK 信号通路在香烟诱导气道上皮细胞黏液高分泌中的作用

    Objective To explore the role of ROS/ Src / JNK signaling pathway in cigarette smoke extract( CSE) -induced mucin ( MUC) 5AC production in A549 airway epithelial cells. Methods The A549 airway epithelial cells were cultured in medium with CSE, then treated with ROS scavenger DMTU, c-Jun Nterminal kinase( JNK) specific inhibitor SP600125, and Src kinase inhibitor PP2, respectively. The relative content of reactive oxygen species( ROS) were assayed by special kit. The levels of MUC5AC in culture medium, epidermal growth factor receptor( EGFR) , activated EGFR and MUC5AC mRNA in culture cells were detected with ELISA,Western Blot and RT-PCR, respectively. Results A dose-dependent increasing of ROS production in cells exposed to dilutions of cigarette smoke solution was detected. DMTU inhibited cigarette smoke-induced Src phosphorylation( P lt; 0. 05) . SP600125 reduced the expression of MUC5AC ( P lt; 0. 05) compared with the normal group. The activation of JNK was suppressed by Src specific inhibitor PP2( P lt; 0. 05) . Conclusion ROS/ Src / JNK signal cascade may play a particular role in MUC5AC expression of A549 cells induced by cigarette smoke.

  • Airway Mucus Hypersecretion in Rats Is Inhibited by the Extracts of Pinellia

    Objective To investigate the effects of extracts of Pinellia ( EP) on a rat model of airway mucus hypersecretion induced by LPS. Methods Thirty Wisatr rats were randomly divided into 5 groups, ie. a blank group, a model group, and three EP groups treated with different doses of EP. There were 6 rats in each group. Airway mucus hypersecretion model was established by intratracheally instillation of LPS in the model group and three EP groups. The rats in three EP groups were orally administered with EP at dosages of 10 g/kg, 30 g/kg and 60 g/kg respectively for 4 days. The expression of Mucin 5AC ( MUC5AC) protein in airway was assayed by immunohistochemistry. The mRNA expressions of MUC5AC and Aquaporin-5( AQP-5) in lung tissue were detected by RT-PCR. ELISA technique was performed to detect TNF-αin bronchoalveolar lavage fluid( BALF) . Results LPS significantly stimulated the mRNA and protein expression of MUC5AC in lung and TNF-αlevel in BALF, and inhibited the expression of AQP-5 mRNA in lung. The EP at dosages of 10 g / kg and 30 g/ kg had little effect on mucus hypersecretion. While 60 g/kg of EP could significantly inhibited the expression of MUC5AC, and decreased the release of TNF-α in BALF. The AQP-5 mRNA was also up-regulated by 60 g /kg of EP. The expression of MUC5AC mRNA was positively correlated with level of TNF-α( r = 0. 948, P lt;0. 05) ; AQP-5 mRNA was negatively correlated with MUC5AC mRNA and TNF-α( r = - 0. 955, P lt; 0. 05; r = - 0. 909, P lt; 0. 05) . Conclusion EP ( 60 g/ kg) can significantly attenuated airway mucus hypersecretion in rats.

  • Association and Research Advancements of Clinical Application in Thomsen-Friedenreich Related Antigens and MUC1 Antigen

    Objective Mucin 1 (MUC1)and Thomsen-Friedenreich related antigens (TFRA)gene play an important role in the occurrence and development of tumor,so we will discuss its research advancements and clinical applications below. Methods Foreign and native related literatures published in recent 10 years were retrieved,and a further exploration on the expression relationship and clinical research progression of MUC1 and TFRA were reviewed. Results MUC1 and TFRA expressed in a high degree of specificity in malignant tumors,and their expressions interacted with each other. Many MUC1 related vaccines had been introduced to clinical in recent years,and some drugs based on TFRA had been put into clinical experiment as gene therapy methods too. Conclusions MUC1 and TFRA may be used as new target antigens of specific immunotherapy for malignant tumor. That they express in high or low level is closely related to tumor biological behavior and prognosis, moreover, they are important to the diagnosis and treatment of cancer.

  • Analysis of Diagnosis and Treatment of Pancreatic Cystic Tumors

    Objective To discuss the methods of diagnosis and treatment of cystic neoplasms of pancreas. Methods Demographic data, clinical manifestations, diagnostic exams, surgical procedures, pathological diagnosis, postoperative complications, and follow-up data of 29 patients with cystic neoplasms of pancreas were analyzed retrospectively. Results There were 8 (28%) serous cystic tumors (SCN), 12 (41%) mucinous cystic tumors (MCN), 3 (10%) intraductal papillary mucinous tumors (IPMN), and 6 (21%) solid pseudopapillary tumors (SPT). Eight cases of SCN, 7 cases of MCN, 1 case of IPMN, and 5 cases of SPT were all benign. The ages of the patients were from 15 to 78 years〔average, (49±17)years〕and all tumors were more common in female (76%, 22/29). Twenty-three cases of 29 patients were performed operations, 22 cases were underwent surgical resection, and 1 case was performed exploration and biopsy. There was no surgery-related death. The rest 6 cases were not performed operation. Twenty-one cases followed-up for 6 months to 8 years 〔average, (2.7±2.3) years〕, 8 cases didn’t followed-up. Sixteen cases with surgical resection had no recurrence during follow-up period, 1 case performed exploration and biopsy died in 1 year after operation, and 4 cases of SCN without surgery didn’t deteriorate. Conclusions The most common cystic neoplasms of pancreas are mucinous and serous cysts. These tumors are more frequent in female. Although almost all serous cysts are benign, 42% of mucinous cysts are malignant. Misdiagnosis may delay appropriate treatment and increase mortality. The resection rate of pancreatic cystic tumor is high, and the prognosis is good after radical resection.

  • Mucinous Versus Nonmucinous Gastric Cancer: Differentiation with 64 Multidetector CT

    Objective To evaluate the capability of 64 multidetector CT in the differentiation between mucinous and nonmucinous gastric cancer. Methods From June 2006 to June 2007, 68 patients diagnosed as gastric cancer (18 mucinous and 50 nonmucinous cancer) underwent preoperative scan with a 64-slice helical CT scanner at West China Hospital. The CT images were analyzed retrospectively on tumor location (proximal/distal stomach), diameter of tumor, appearance of thickened gastric wall, contrast enhancement pattern (layered/nonlayered), degree of enhancement and serosal invasion. Results The primarily thickened layer (94% of patients) was the low attenuation middle layer in mucinous cancer and the rate was 72% with high attenuating inner layer or entire layer in nonmucinous cancer. The most common contrast enhancement pattern and degree was layered (83% of patients) and non-enhanced (89% of patients) in mucinous cancer and nonlayered (76% of patients) with enhanced (60%of patients) in nonmucinous cancer. The more common serosal invasion was shown in mucinous cancer than in nonmucinous cancer (89% vs 64%). These findings were statistically significant (P<0.05). Tumor location and size of gastric cancer were not correlated with operative pathologic classification. Conclusion Sixty-four multidetector CT is effective in distinguishing mucinous from nonmucinous gastric cancer, predominantly on the basis of thickened and layered gastric wall, enhancement pattern of low-attenuating middle layer.

  • Research Progress of Mucinous Breast Cancer

    ObjectiveTo summarize the research progress of mucinous breast cancer (MBC). MethodsLiteratures about the recent studies of MBC were reviewed. ResultsMBC was one of special subtype of infiltrating breast cancer. According to the mucus ingredient in the ratio of the mass, MBC was divided into pure mucinous breast cancer (PMBC) and mixed mucinous breast cancer (MMBC). Compared to infiltrating ductal cancer-not otherwise specified (IDC-NOS), MBC showed higher positive expression rates of estrogen receptor (ER) and progestrogen receptor (PR), with reduced lymph node metastasis rate and better prognosis. PMBC had lower lymph node metastasis rate and better outcome than MMBC. ConclusionsThere is significant difference about clinical and pathological characteristics between MBC and IDC-NOS. Researches are generally believed that MBC is an uncommon breast neoplasm which is associated with a good prognosis.

  • The effects of elafin on the regulation of mucin5AC in airway epithelial cells

    ObjectiveTo explore the effects of elafin on the regulation of mucin5AC (MUC5AC) in airway epithelial cells.MethodsAfter cultivating epithelial cells in vitro, cells were stimulated by cigarette smoke extract (CSE) and lipopolysaccharide (LPS), the intracellular MUC5AC production and extracellular secretion were assayed by immunofluorescence and ELISA, respectively.ResultsThe level of MUC5AC protein was obviously increased in the LPS stimulation group [(0.785±0.005) μg/ml vs. (0.232±0.004) μg/ml] and the CSE stimulation group [(0.803±0.005) μg/ml vs. (0.255±0.003) μg/ml] than those in the blank control groups (both P<0.01). When pretreated with elafin then stimulated by LPS and CSE, the MUC5AC productions were evidently decreased than those in the single LPS and CSE stimulation groups [(0.363±0.003) μg/ml and (0.406±0.006) μg/ml, bothP<0.01]. As compared with the blank control groups, the MUC5AC protein secretion were significantly decreased after stimulated with single elafin [(0.176±0.002) μg/ml and (0.193±0.004) μg/ml, bothP<0.05].ConclusionElafin may inhibit MUC5AC hypersecretion by blocking MUC5AC production and secretion.

  • Expression and pathological significance of neutrophil elastase and mucin 5AC in rats with chronic obstructive pulmonary disease

    ObjectiveTo investigate the expression and pathological significance of neutrophil elastase (NE) and mucin 5AC (MUC5AC) in chronic obstructive pulmonary disease (COPD) rats. MethodsA total of 60 male specific pathogens free Sprague-Dawley rats were randomly divided into model group and blank group, each with 30 rats. The number of goblet cells and the positive expression of NE and MUC5AC were compared between the two groups by hematoxylin-eosin staining, alcianblue-periodic acid Schiff staining and immunohistochemical staining. ResultsNo rats died accidentally during the experiment. The number of goblet cells was 57.93±7.79 in the model group and 6.80±3.19 in the blank group, and the difference between the two groups was statistically significant (t=−44.267, P<0.05). The median (lower quartile, upper quartile) of the positive expression score of NE was 0 (0, 0) in the blank group and 2 (2, 3) in the model group, the median (lower quartile, upper quartile) of the positive expression score of MUC5AC was 0 (0, 1) in the blank group and 2 (2, 2) in the model group, and the differences between the two groups were statistically significant (P<0.05).ConclusionsThe positive expression of NE, goblet cells, MUC5AC and mucus secretion in COPD model rats increases significantly. Perhaps NE and MUC5AC can be used as target interventions to inhibit mucus hypersecretion.

  • The effects of astaxanthin on the airway inflammation and remodeling in the asthmatic rats

    Objective To observe the effects of astaxanthin (AST) on the airway inflammation and remodeling in the asthmatic rats. Methods Fifty male Wistar rats were randomly divided into five groups (n=10 for each group): saline-sensitized and-saline-challenged group (the control group), bronchial asthma group (the asthma group), bronchial asthma+astaxanthin 5 mg/kg gavage treatment group (the AST 5 mg/kg group), bronchial asthma+10 mg/kg gavage treatment group (the AST 10 mg/kg group), and bronchial asthma+50 mg/kg gavage treatment group (the AST 50 mg/kg group). The level of interleukin-5(IL-5), interleukin-13(IL-13), interferon-γ(IFN-γ), tansforming growth factor-β (TGF-β), malondialdehyde (MDA) and superoxide dismutase (SOD) in the bronchoalveolar lavage fluid (BALF) and the total IgE level in the serum were measured using enzyme linked immunosorbent assay (ELISA).The infiltration of airway inflammatory cells and the degree of airway epithelial cells detachment, the extent of goblet cell hyperplasia and the severity of subepithelial collagen deposition were evaluated on the hematoxylin eosin (HE), periodic acid Schiff (PAS) and Masson trichrome stained lung sections. reverse transcription-polymerase chain reaction (RT-PCR) was used to measure the expression of mucin 5A and C (MUC5AC) messenger ribonucleic acid(mRNA) in lung tissue; Immunohistochemical staining was used to determine the expression of MUC5AC protein in the rat airway epithelium. Results The level of IL-5, IL-13, TGF-β, MDA and the total IgE in the serum respectively [(36.73±2.29), (53.99±2.70), (60.89±2.54)ng/mL,(18.65±0.76)umol/L, (54.50±2.91)ng/mL], the extent of inflammatory cells infiltration (46.24 ± 4.26), the extent of eosinophils infiltration (2.09± 0.13), the extent of epithelial cells detachment [(6.09±0.45)%], the extent of goblet cell hyperplasia [(13.65±1.90)%], the extent of subepithelial collagen deposition [(17.58±2.14)%], the MUC5AC mRNA expression level, and the lung tissue MUC5AC protein expression IOD value (187±12) in the asthma group were all higher than those in the control group (P<0.01 or P<0.001), the level of IFN-γ and SOD in the BALF[(26.38±1.70) ng/mL], [(16.37±1.22) U/L], was lower than that in the control group (P<0.001); The level of IL-5, IL-13, total IgE, TGF-β, MDA, the inflammatory cells infiltration in the airway epithelial, the degree of epithelial cell damage and detachment, the degree of goblet cell hyperplasia, the degree of subepithelial collagen deposition, the MUC5AC mRNA expression in lung tissue,and the MUC5AC protein expression in airway epithelial cells in the AST treated groups were all lower than those in the asthma group (P<0.05 or P<0.01 or P<0.001),the level of IFN-γ, SOD in the BALF was higher than that in the asthma group (P<0.05 or P<0.01). Conclusion Astaxanthin can inhibit airway inflammation, downregulate airway MUC5AC expression, inhibit goblet cell proliferation, and alleviate airway remodeling in rats with bronchial asthma.
