  • Effect of aerosolized perfluorocarbon on the tumor necrosis factor-α mRNA expression and lung histopathology in rabbits with acute lung injury

    Objective To investigate the effect of aerosolized perfluorocarbon (PFC) (FC77) on gas exchange,histopathological changes of lung in acute lung injury and pulmonary expression of tumor necrosis factor-α (TNF-α) mRNA.Methods After acute lung injury (ALI) was induced by oleic acid (OA),16 rabbits were assigned randomly into 2 groups,ie.aerosolized perfluorocarbon group (PFC group) and conventional mechanical ventilation group (CMV group).Gas exchange parameters were measured before and after ALI,at 1,2,3,4 h after treatment.Histological sections taken from 6 different parts of lung were stained by hematoxylin and eosion.The express of TNF-α mRNA in the 2 different parts of lung were detected by in situ hybridization (ISH).Results Compared with CMV group,the PaO2 and static lung compliance (CLst) were significantly increased (Plt;0.05),the histopathological lesions of lung were attenuated,and the TNF-α mRNA expression was decreased significantly in PFC group (all Plt;0.05).There was more expression of TNF-α mRNA in backside than that in foreside of lung in two groups (Plt;0.05).Conclusion Aerosolized perfluorocarbon (PFC) can decrease expression of tumor necrosis factor-α mRNA in the lung,and improve the CLst and oxygenation during acute lung injury.

  • Safety study of intravenous infusion with self-made perfluorocarbon emulsion in rats

    Objective To assess the safety of intravenous infusion with self-made perfluorocarbon emulsion (PFE) in rats.Methods 35 Wistar rats were randomly divided into a normal control group and six PFE groups (n=5 in each group).The control group was intravenous injected with normal saline and six PFE groups with PFE.Blood samples were obtained from the PFE groups 5 min after injection via vena orbitalis for perfluorocarbon (PFC) measurement.The control group were sacrificed at 2 hours after injection,and the six PFE groups were respectively killed at 2 h,4 h,6 h,24 h,48 h and 10 d after injection.The blood levels of PFC,PaO2,GPT,GOT,BUN and Cr were measured.Pathological changes of lung,liver and kidney were also observed.Results All rats were in good health state after PFE intravenous injection with no obvious pathological changes in liver and kidney.PFC was wide distributed throughout the pulmonary alveoli and capillaries.The levels of GPT and GOT were higher in the PFC groups at 2,4,6 and 24 h than which in the control group (all Plt;0.05),but there were no significant difference between the PFE 10 d group and the control group.The levels of BUN and Cr had no remarkably differences in all groups.Blood PFC concentrations were (20±1.8)mg/mL,(1.8±0.7)mg/mL,(1.5±0.6)mg/mL,(1.2±0.4)mg/mL,(0.5±0.2)mg/mL,(0.2±0.03)mg/mL,0 mg/mL in the PFE groups respectively at 5 min,2 h,4 h,6 h,24 h,48 h,10 d after injection.PaO2 of the PFE 2 h group was significant higher than which in the control group [(119.2±8.6)mm Hg vs (99.6±4.7)mm Hg,Plt;0.05].Conclusion Intravenous injection with PFE (10 mL/kg) can enhance PaO2 with good safety and no obvious influence on pathology of lung,liver and kidney,so may be used for the treatment study of acute lung injury.

  • Effects of Vaporized Perfluorocarbon Inhalation on Histopathology of Lung and Extra-Pulmonary Organs in Rabbits with Acute Lung Injury

    Objective To investigate the effects of vaporized perfluorocarbon( PFC) inhalation on histopathology of lung, small intestine, liver and kidney of acute lung-injured rabbits. Methods Eighteen New Zealand rabbits were randomly divided into 3 groups, ie. a conventional mechanical ventilation( CMV)group, a PFC group, and a control group. The rabbits were mechanical ventilated and intratracheally infused artificial seawater to induce acute lung injury. After ALI was established( PaO2 /FiO2 lt; 200 mm Hg) , the CMV group received CMV for 6 hours. The PFC group received PFC inhalation for 2 hours, and followed by CMV for 4 hours. And the control group was weaned from ventilation. Then they were sacrificed for histopathological measurement of lung, small intestine, liver and kidney. Results The rabbits in the control group died in 15 minutes after discontinuation of ventilation. Vaporized PFC inhalation can obviously improve oxygenation and attenuate the damage of the lung in contrast to CMV. Mild improvement was observed in small intestine, liver and kidney after vaporized PFC inhalation, but without statistical significance. Conclusion Vaporized PFC inhalation can improve oxygenation and attenuate lung injury in histopathology,but have no apparent protective effects on extra-pulmonary organs.

  • Experimental Research of Partial Liquid Ventilation with Perfluorocarbon on Acute Lung Injury Induced by Cardiopulmonary Bypass

    Objective To investigate the effect of partial liquid ventilation (PLV) with perfluorocarbon(PFC) and continuous pulmonary artery perfusion (CPP) on lung gas exchange and lung inflammatory reaction in acute lung injury(ALI) induced by cardiopulmonary bypass (CPB). Methods Eighteen of either sex piglets(weighting10.2±1.6kg) were randomly divided into three groups: Control group, CPP+CPB group (CPP group), PLV+CPP+CPB group (PLV group). Animals in control group received no treatment but conventional mechanical ventilation.In CPP group lung perfusion with oxygenated blood at 20-25ml/kg·min was given during aortic clamping. In PLV group PFC (FDC)12ml/kg was instilled into the trachea right after CPB stopping. The changes of gas exchange were mearsured before CPB and at 0h, 1h, 2h, 3h after CPB stopping. Histological sections were taken from right and left downsides of lung. Results Compared with control group, the partial pressure of oxygen in artery (PaO2) significantly increased and alveolar-aterial oxygen gradient(AaDO2) markedly decreased after 1h in PLV group(Plt;005) and partial pressure of carbon dioxide in artery (PaCO2) also became small after 3h (Plt;005).The change of gas exchange in CPP group was markedly improved. And role of lung protection of PLV was more better than that of CPP. Light microscopy: Express of intercellular adhesion molecule-1(ICAM-1) in the histopathological lesions of lung was bely positive in control group than that of PLV group and CPP group. Conclusion PLV and continuous pulmonary artery perfusion can improve the oxygenation of lung and inhibit inflammatory reaction of acute lung injury induced by CPB
