  • Relationship Between cmet, p53 Expression in Breast Carcinoma and Tumor Angiogenesis

    ObjectiveTo find the role of oncogene cmet and suppressor gene p53 in the process of tumor angiogenesis and their clinical significance. MethodsBy immunohistochemical method and computer image analysis technique, microvessel count and cmet, p53 protein expression were quantitatively determined in 80 cases of breast carcinoma and 20 cases of breast fibroadenoma. ResultsThe high microvessel count and the positive expression of cmet, p53 were significantly correlated with histologic grade, lymph node metastasis and the stage of the tumor (P<0.01). The high microvessel count was significantly correlated with the positive expression of cmet and p53 (P<0.01).ConclusionBoth oncogene cmet and suppressor gene p53 modulate tumor angiogenesis of breast carcinoma.

  • Expression of hepatocyte growth factor receptor in human epiretinal membranes and RPE cells

    Objective To investigate the expression of hepatocyte growth factor receptor (HGFR) in epiretinal membranes (ERM) of eyes with proliferative vitreoretinopathy (PVR) and cultured retinal pigent epithelium (RPE) cells. Methods Fifteen human epiretinal membranes were obtained from eyes undergone vitrectomy for rhegmatogenous retinal detachment complicated with PVR and observed by immunohistochemical examination to study the expression of HGFR. Using the immunohistochemical technique to evaluate the expression of HGFR in cultured RPE cells. Results In 6 membranes of PVR-grade C, HGFR were expressed in 5/6, and 7 cases were detected in 9 membranes of PVR-grade D.RPE cells express readily detectable levels of HGFR. Conclusion The findings indicate that HGF might be involved in the formation of epiretinal membranes in PVR. (Chin J Ocul Fundus Dis, 2002, 18: 221-223)

  • Expression of HGF and c-met Detected by Tissue Microarray and Relationship with Tumor Angiogenesis in Human Colorectal Cancer

    Objective To study hepatocyte growth factor (HGF) and its receptor (c-met) expressions in human colorectal cancer and non-neoplasm colorectal mucosa, and the relationship with tumor angiogenesis. Methods Tissue microarrays (TMAs) were made up of 80 cases of colorectal cancer and 80 cases of nonneoplasm colorectal mucosa. The expressions of HGF and c-met were detected by immunohistochemistry (SP). CD105 was used as a marker to account microvessel density (MVD) in tumor tissue. Results HGF was over expressed in 48 cases and c-met was over expressed in 63 cases of colorectal cancer tissue, and the correlation between HGF and c-met positive expression was significant (r=0.231, Plt;0.05). The high expression rate of HGF and cmet in colorectal cancer were significantly higher than that in non-neoplasm colorectal mucosa (χ2=35.387, Plt;0.05; χ2=59.854, Plt;0.05) of colorectal cancer. The overexpression of HGF was correlated with lymph node metastasis (χ2=4.743, Plt;0.05) and TNM staging (χ2=5.576, Plt;0.05). The overexpression of c-met was correlated with differentiation (χ2=15.767, Plt;0.05) and lymph node metastasis (χ2=5.765, Plt;0.05) of colorectal cancer. MVD was different between overexpression and lowexpression colorectal cancer tissues of HGF and cmet (t=2.150, Plt;0.05; t=2.052, Plt;0.05). There was statistical correlation between HGF and cmet overexpression (r=0.259, Plt;0.05). The overexpressions of HGF and cmet were correlated with lymph metastasis in moderate differentiation cancer (χ2=13.154, Plt;0.05; χ2=5.371, Plt;0.05). Conclusions The overexpressions of HGF and c-met in colorectal cancer may be related with tumor angiogenesis. Detecting the expressions of HGF and c-met is valuable to estimate the biological character of colorectal cancer.

  • c-met Expression in Tall Cell Variant Papillary Carcinoma of The Thyroid

    Objective To investigate the expression of c-met in tall cell variant of papillary thyroid carcinoma, and to compare it with other types of thyroid carcinoma and benign thyroid tissue. Methods The expressions of c-met in 60 cases of thyroid specimens were tested by immunohistochemical staining. Results The levels of expressed c-met in tall cell variant specimens were significantly higher than those in other types of papillary thyroid carcinoma and benign thyroid tissue. c-met expressions were significantly different in the following pairs of types: tall cell variant vs common papillary carcinoma of thyroid (P=0.000 1), tall cell variant vs follicular variant papillary thyroid carcinoma (P=0.000 1), and tall cell variant vs benign thyroid tissue (P=0.000 1). In addition, for all types of papillary carcinomas evaluated, c-met expression was significantly higher in specimens with extracapsular spread (P=0.010 0) and skeletal muscle invasion (P=0.020 0). Conclusion The high expression of c-met is a significant marker for tall cell variant papillary carcinoma of thyroid and its invasive behavior. This finding may explain the unusually aggressive behavior of this tumor and suggest a role for c-met in the early identification of patients with tall cell variant thyroid carcinoma.

  • Expression of c-met mRNA in The Drainage from Axillary Fossa after Operation for Breast Cancer

    Objective To study the significance of c-met mRNA in axillary drainage after operations for breast cancer. Methods RT-PCR assay was used to examine c-met mRNA in axillary drainage after operations in 52 cases of breast cancer. The relationships between the expression of c-met and the tumor size, metastatic lymph nodes, the expressions of estrogen receptor (ER), progesterone receptor (PR) and c-erbB-2 were analyzed, respectively. In addition, the effect of douching operative field with 5-FU and distilled water on the expression of c-met mRNA was also analyzed. Results ①The proto-oncogene c-met mRNA could be detected in axillary drainage after operations for breast cancer by RT-PCR, and its positive rate was higher than that in routine pathological detection for micrometastasis in the axillary lymph nodes (P<0.05). ②The expression of c-met mRNA was correlated with both the metastatic lymph nodes and tumor size. ③There was no significant relationship between the expression of c-met mRNA and the expressions of ER, PR and c-erbB-2. ④Dounching operative field with 5-FU and distilled water could decrease the expression of c-met mRNA.Conclusion The proto-oncogene c-met mRNA may be an ideal and specific marker for dectecting micrometastasis of breast cancer. In addition, it also suggests that the examination of c-met mRNA in the axillary drainage by RT-PCR assay could detect the micrometastasis in axillary lymph nodes much easier and more accurately than routine pathological method.

  • THE ADVANCE IN RESEARCHING INTO HEPATOCYTE GROWTH FACTOR AND RECEPTOR c-met IN BREAST CARCINOMA

    Objective To study the effects of hepatocyte growth factor (HGF) and receptor c-met on the development of primary breast carcinoma, and the relationship between it and prognosis. Methods The study of HGF and c-met related to breast carcinoma was reviewed by history document and experimental study in recent years. Results HGF is a growth factor which has mitogenic, migrating, invasive and angiogenic activities in breast carcinoma cells. The carcinogenic mechanism of breast carcinoma was more clear with the discovery of the relationship between HGF and its receptor c-met. Conclusion The HGF/c-met plays an important role in the generation and progress of breast carcinoma. Studying the effects of HGF/cmet on breast carcinoma is significant in guiding clinical treatment.

  • THE EXPRESSIONS AND SIGNIFICANCE OF C-MET ONCOPROTEIN, TRANSFORMING GROWTH FACTOR-α IN BENIGN AND MALIGNANT LESIONS OF BREAST

    The expressions and significance of c-met oncoprotein and transforming growth factor-α (TGF-α) were studied by immunohistochemical method in 50 cases of breast cancer (BC) and 12 cases of benign lesions of breast (BL). The positive rate of c-met, TGF-α in BC was 26.0% and 25.0% respectively, in BL was 8.3% and 25.0% respectively. The positive rate of c-met oncoprotein was lower in the cases of histologic Grade Ⅰ, positive of ER and PR or CEA than that of histologic Grade Ⅲ, negative of ER and PR or CEA. The positive rate of TGF-α was lower in the cases of histologic Grade Ⅰ, negative of ER and PR or CEA than that of histologic Grade Ⅲ, positive of ER and PR or CEA. These results suggest the expression of c-met and TGF-α might be related to the carcinogenesis and development or endocrine state of BC.

  • Estimation of Radiation Dose from Patients Undergoing 99mTc-Methoxy Isobutyl Isonitrile Myocardial Perfusion Imaging to the General Public and Nuclear Medicine Technicians

    ObjectiveTo estimate the radiation dose (RD) to the general public and nuclear medicine technicians from patients undergoing 99mTc-methoxy isobutyl isonitrile (MIBI) myocardial perfusion imaging. MethodsFrom January to June 2015, 55 patients including 30 males and 25 females aged between 25 and 87 years[averaging (63.6±15.1)years] ready to undergo myocardial perfusion scintigraphy with 99mTc-MIBI were prospectively recruited in this study. Approximately at hour 1.5 after injection of 99mTc-MIBI, whole-body dose-equivalent rate was measured with a radiation-survey meter at 0.3 meter and 1.0 meter from the patients. On the basis of human 99mTc-MIBI metabolic rate proposed by the International Commission of Radiological Protection and human social contact model proposed by the National Council on Radiation Protection and Measurements, the RDs to the general public from patients who had completed 99mTc-MIBI myocardial perfusion imaging and left nuclear medicine department were calculated. On the assumption that a nuclear medicine technician typically spent 5 minutes at a distance of 0.3 meter for positioning the patient, the technician's RD was also estimated. ResultsThe RD to a family member sleeping with the patient at night was predicted to be 42.88-160.55 μSv, to a family member contacting the patient at daytime 7.50-29.38 μSv, to a colleague 9.89-38.78 μSv and to a nearby passenger 124.48-466.06 μSv. The RD to a technician per 99mTc-MIBI myocardial perfusion imaging procedure was predicted to be 1.72-6.44 μSv. ConclusionThe predicted RDs to the general public and technicians from exposure of patients undergoing 99mTc-MIBI myocardial perfusion imaging are significantly lower than the regulatory dose limits.

    As the incidence of osteoporosis (OP) is increasing year by year, high morbidity and mortality caused by osteoporotic fractures have become major problems of health in China and over the world. Quantitative measurement of bone using 99mTc-methylene diphosphonate (99mTc-MDP) provides global or local information of skeletal metabolism or transformation. In this paper, we make a brief review on the quantitative measurement of bone using 99mTc-MDP and expect to provide guidance for clinical diagnosis and treatment.

  • Bariatric-metabolic surgery and it’s effect on systemic system

    Objective To understand bariatric-metabolic surgery and explore it’s effects on all systems of body. Method The literatures relevant bariatric-metabolic surgery at home and abroad were reviewed and analyzed. Results The bariatric-metabolic surgery continued to evolve, which had undergone the changes of the open and minimally invasive therapeutic pathways, it’s effect of weight reduction was remarkable and had a low incidence of postoperative complications. But there was still no " gold standard” for selection of operation method. The bariatric-metabolic surgery affected the changes of all the systems of the body, could cure or improve the obesity related diseases. Conclusions With minimal invasion and precision of bariatric-metabolic surgery, although it’s complications is less and less, there are still some problems. We need to fully evaluate by multidisciplinary, pay attention to physiological and psychological factors, adopt scientific nutrition management, promote benign development of bariatric-metabolic surgery.
